熊果酸对顺铂耐药人卵巢癌SKOV3/DDP细胞增殖与凋亡的影响

摘    要:目的探讨熊果酸对体外培养的顺铂耐药人卵巢癌SKOV3/DDP细胞增殖与凋亡的影响。方法将熊果酸作用于体外培养的顺铂耐药人卵巢癌SKOV3/DDP细胞,采用MTT法检测不同浓度的熊果酸(0、10、20、40、50、60、80μmol.L-1)对SKOV3/DDP细胞生长的抑制作用,采用流式细胞术检测SKOV3/DDP的细胞凋亡和细胞周期。结果不同浓度的熊果酸对体外培养的SKOV3/DDP细胞均有明显的抑制增殖作用,并呈剂量、时间依赖效应(均P〈0.05),熊果酸(60μmol.L-1)作用于SKOV3/DDP细胞48 h后,达到最大细胞凋亡率(31.22±1.98)%;熊果酸可影响SKOV3/DDP细胞增殖周期中的G0/G1期,使细胞在此期停滞,诱导其发生凋亡。结论熊果酸对体外培养的顺铂耐药人卵巢癌SKOV3/DDP细胞增殖有抑制作用,并能诱导细胞凋亡。

Abstract:Objective To investigate the effects of ursolic acid(UA) on proliferation and apoptosis of drug-resistant SKOV3/DDP ovarian carcinoma cells in vitro.Methods Drug-resistant SKOV3/DDP ovarian carcinoma cells were treated with different concentrations of UA(0,10,20,40,50,60 and 80 μmol·L-1).MTT assay was performed to detect cell proliferation,and flow cytometry was used to determine cell cycle and apoptosis.Results UA markedly inhibited the proliferation of SKOV3/DDP ovarian carcinoma cells in a dose-and time-dependent manner(P〈0.05),and induced G0/G1 cell cycle arrest and apoptosis.The higher concentration of UA(60 μmol·L-1) resulted in the highest apoptosis rate(31.22±1.98)%] after treatment for 48 hours.Conclusion UA can inhibit cell proliferation and induce apoptosis in drug-resistant SKOV3/DDP ovarian carcinoma cells.
