摘    要:目的观察复方小柴胡汤(FFXCHT)对高分化鼻咽癌细胞CNE-1和低分化鼻咽癌细胞CNE-2增殖的抑制作用,探讨其调控CNE-2细胞microRNA的差异表达。方法通过磺基罗丹明B(SRB)法检测FFXCHT对CNE-1和CNE-2细胞增殖的影响;采用μParaflo^TM MicroRNA芯片检测,激光扫描器收集杂交的图像,经LOWESS滤器规范信号后比较数据,分析FFXCHT处理前后CNE-2细胞microRNA表达的差异。结果FFXCHT可抑制CNE-1和CNE-2细胞增殖,且呈剂量依赖性;FFXCHT处理CNE-1细胞24h、48h、72h,其IC50分别为12.23mg/mL,4.63mg/mL和4.97mg/mL;其对CNE-2细胞IC50分别为9.67mg/mL,4.33mg/mL,3.09mg/mL。microRNA芯片检测结果显示FFXCHT处理CNE-2细胞后,在检测的326个microRNA中,有10个microRNA上调,1个microRNA下调,其中检测量绝对值在1000以上且两者相差log2(Sample B signal/Sample A signal)〉1的microRNA有hsamiR-513、hsa-miR-498、hsa-miR-210和hsa—miR-602。结论该FFXCHT可抑制高低分化鼻咽癌细胞的增殖,其抑瘤作用可能与microRNA的差异表达有关。

Abstract:Objective To investigate the effect of a new complex traditional Chinese medicine(TCM) on the proliferation of well-differentiated and poorly differentiated nasopharyngeal carcinoma cells, CNE-1 and CNE-2, as well as the effect of it on the expression of microRNA in CNE-2. Methods The effect of complex TCM on proliferation of CNE-1 and CNE-2 were detected by Sulforhodamine B(SRB) assay. MicroRNA were detected by μParaflo microfluidic microRNA chip, and hybridization images collected using a laser scanner were normalizing using a LOWESS filter to analysis the microRNA expression discrepancy of CNE-2 treated by complex TCM. Results Complex TCM can dose-dependently inhibit proliferation of CNE-1 and CNE-,2; Comparing to control, 10 microRNA were up-regulation and 1 microRNA was down-regulation in CNE-2 treated by complex TCM among 326 microRNA detected, of which absolute value of detection〉1000 and log2 (Sample B signal/ Sample A signal)〉1 was hsa-miR-513, hsa-miR-498, hsa-miR-210 and hsa-miR-602. Conclusion Complex TCM can inhibit proliferation of CNE-1 and CNE-2; The effect may be related to take part in the microRNA regulation of nasopharyngeal carcinoma cells.
