摘    要: 目的 探讨去甲斑蝥素(NCTD)对人类胃癌SGC-7901细胞凋亡相关基因表达的影响。方法 实验分对照组、5-Fu组、NCTD 组和5-Fu+NCTD 组,分别应用MTT、流式细胞术、免疫组化SABC法检测NCTD 对体外培养胃癌SGC-7901细胞的杀伤抑制率、凋亡率和survivin、bcl-2、caspase-3的表达。结果 NCTD对人类胃癌SGC-7901 细胞生长有抑制作用,呈剂量- 时间效应关系;凋亡率由(8.30±1.49)%上升到(20.56±1.32)%;survivin基因表达由(86.57±4.39)%下降到(26.11±2.27)%;bcl-2基因表达由(85.35±3.25)%下降到(30.26±1.83)%;caspase-3基因表达由(54.49±3.07)%上升到(92.78±2.47)%;与5-Fu联合应用上述作用更明显。结论 NCTD能抑制人类胃癌SGC-7901细胞的生长;与5-Fu具有协同作用,其机制可能与其诱导细胞凋亡,影响凋亡相关基因表达有关。

Abstract:Objective To investigate the influence of Norcantharidin (NCTD) on apoptosis-related gene expression of gastric cancer cell line SGC-7901. Methods The experiment was divided into control group, 5-Fu group, NCTD group and 5-Fu+NCTD group. The inhibitory rate, apoptosis rate and expression of survivin, bcl-2, and caspase-3 were detected by MTT assay, flow cytometry and SABC immunohistochemical method, respectively. Results NCTD showed that the inhibitory effect on growth of SGC-7901 cells with a dose -and time-effective dependent manner. The apoptotic rate increased from (8.30±1.49)% to(20.56±1.32)%. The expressions of survivin decreased from (86.57±4.39)% to (26.11±2.27)% and bcl-2 from (85.35±3.25)% to (30.26±1.83)%, while caspase-3 increased from (54.49±3.07)% to (92.78±2.47)%, 5-Fu had the synergistic effects with NCTD. Conclusion NCTD can inhibit the growth of SGC-7901 cell lines. 5-Fu had the synergistic effects with NCTD. The mechanism might correlate with induction of cell apoptosis via effect of the expression of apoptotic-related genes such as survivin, bcl-2 and caspase-3.
