miR-34在胃癌组织中的表达及与EMT、化疗耐药性的相关性

摘    要:目的:探究微小RNA-34(miR-34)在胃癌组织中的表达及与上皮-间质转化(EMT)、化疗耐药性的相关性。方法:选取2017年3月-2018年3月于我院接受治疗的胃癌患者50例,取手术切除癌组织以及癌旁组织标本待用。Western blot法检测miR-34及EMT相关蛋白上皮性钙黏蛋白(E-cadherin)、波形蛋白(Vimentin)、snail相对表达量。根据化疗耐药性将所有患者分为化疗耐药组、化疗敏感组,对两组患者化疗前胃癌组织中miR-34、E-cadherin、vimentin、snail的相对表达量进行对比。结果:miR-34、E-cadherin在胃癌组织中的表达显著低于癌旁组织(P<0.05);vimentin、snail在胃癌组织中的表达显著高于癌旁组织(P<0.05)。病理分期Ⅲ、Ⅳ期患者miR-34、E-cadherin表达低于病理分期Ⅰ、Ⅱ期患者,vimentin、snail表达高于病理分期Ⅰ、Ⅱ期患者(P<0.05);有淋巴结转移患者miR-34、E-cadherin表达低于无淋巴结转移患者,vimentin、snail表达高于无淋巴结转移患者(P<0.05);肿瘤体积≥5 cm3患者miR-34、E-cadherin表达低于肿瘤体积<5 cm3患者,vimentin、snail表达高于肿瘤体积<5 cm3患者(P<0.05);低分化患者miR-34、E-cadherin表达低于高分化、中分化患者,vimentin、snail表达高于高分化、中分化患者(P<0.05)。miR-34表达与E-cadherin表达在胃癌中呈正相关,miR-34表达与vimentin、snail表达在胃癌中呈负相关。化疗敏感组患者化疗前miR-34、E-cadherin表达低于化疗耐药组,vimentin、snail表达高于化疗耐药组(P<0.05)。结论:miR-34、E-cadherin、vimentin、snail在胃癌组织中呈现异常表达,且具有一定的相关性,且miR-34、E-cadherin、vimentin、snail表达与化疗耐药性具有一定的相关性。

Abstract:Objective:To investigate the expression of microRNA-34 (miR-34) in gastric cancer cells and its correlation with epithelial mesenchymal transition (EMT) and chemoresistance.Methods:Fifty patients with gastric cancer who were treated in our hospital from March 2017 to March 2018 were selected,and specimens of resected cancer tissues and adjacent tissues were taken for future use.Western blot was used to detect the relative expression of miR-34 and EMT-related proteins,such as E-cadherin,Vimentin and snail.According to chemoresistance,all patients were divided into chemoresistance group and chemosensitivity group,and the relative expression levels of miR-34,E-cadherin,vimentin and snail in gastric cancer tissues before chemotherapy were compared between the two groups.Results:The expression of miR-34 and E-cadherin in gastric cancer tissues was significantly lower than that in adjacent tissues (P<0.05).The expression of vimentin and snail in gastric cancer tissues was significantly higher than that in adjacent tissues (P<0.05).The expression of miR-34 and E-cadherin in patients with pathological stage III and IV was lower than that in patients with pathological stage I and II,and the expression of vimentin and snail was higher than that in patients with pathological stage I and II (P<0.05).The expression of miR-34 and E-cadherin in patients with lymph node metastasis was lower than that in patients without lymph node metastasis,and the expression of vimentin and snail was higher than that in patients without lymph node metastasis (P<0.05).The expression of miR-34 and E-cadherin expression in tumor volume≥5 cm3 was lower than patients with tumor volume<5 cm3,vimentin and snail expression was higher than patients with tumor volume<5 cm3 (P<0.05).The expression of miR-34 and E-cadherin was lower in poorly differentiated patients than in well-differentiated and moderately differentiated patients,and vimentin and snail expression was higher than well-differentiated and moderately differentiated patients (P<0.05).The expression of miR-34 was positively correlated with E-cadherin expression in gastric cancer,while the expression of miR-34 was negatively correlated with vimentin and snail expression in gastric cancer.Before chemotherapy,the expression of miR-34 and E-cadherin in chemosensitive group was lower than that in chemoresistant group,and the expression of vimentin and snail was higher than that in chemoresistant group (P<0.05).Conclusion:The expression of miR-34,E-cadherin,vimentin and snail were abnormal and correlated in gastric cancer,and the expression of miR-34,E-cadherin,vimentin and snail was correlated with chemotherapy resistance. 【Key words】 Modern Oncology 2022,30(16):2952-2956
