摘    要:目的:探讨22RV1型前列腺癌荷瘤裸鼠模型的制作,以及18F-FPA在裸鼠模型中的显像价值。方法:选取22RV1型前列腺癌细胞株进行培养,饲养BALB/c型裸鼠10只,将指数生长期细胞接种于裸鼠右前臂皮下,定期观察成瘤情况。利用CFN-200多功能模块合成显像剂18F-FPA,然后对荷瘤裸鼠进行PET/CT显像,并与18F-FDG和11C-胆碱显像进行对比,评价18F-FPA对前列腺癌的显像价值。结果:BALB/c型裸鼠一次接种的成瘤率为80%(8/10),经一次补种后成瘤率达100%(10/10)。18F-FPA的合成过程约40 min,放射化学纯度大于97%,符合显像要求。静脉注射显像剂18F-FPA后0.5 h、1 h、2 h,肿瘤均能够清晰显像,最大标准化摄取值(SUVmax)分别为5.79±0.78、6.22±0.95、6.29±0.94,无明显统计学差异。对比显像显示,肿瘤对18F-FDG和11C-胆碱仅有轻微摄取,SUVmax均小于18F-FPA显像(P<0.01)。结论:用22RV1型前列腺癌细胞株和BALB/c裸鼠可成功制作前列腺癌荷瘤裸鼠模型。 18F-FPA在22RV1型前列腺癌荷瘤裸鼠模型上有良好的显像效果,但还需要和其他新型显像剂做进一步对比研究。

Abstract:Objective:To investigate the establishment of 22RV1 tumor bearing nude mouse model of prostate cancer and the imaging value of 18F-FPA.Methods:22RV1 prostate cancer cell lines were selected for culture,and 10 BALB/c nude mice were raised.The exponential growth period cells were inoculated under the skin of the right forelimb of nude mice,and tumor formation was regularly observed.The imaging agent 18F-FPA was synthesized by CFN-200 multifunctional module,and then the imaging of tumor-bearing nude mice was performed,and compared with 18F-FDG and 11C-choline imaging,to evaluate the imaging value of 18F-FPA for prostate cancer.Results:The tumorigenesis rate was 80%(8/10)after the first inoculation and 100%(10/10)after the reseeding.The synthesis process of 18F-FPA was about 40 min,and the radiochemical purity was over 97%,which met the imaging requirements.Tumor imaging could be clearly developed at 0.5 h,1 h and 2 h after intravenous injection of imaging agent 18F-FPA,and the maximum standardized uptake values(SUVmax)were 5.79±0.78,6.22±0.95 and 6.29±0.94 respectively,showing no significant statistical difference(P>0.05).Contrast imaging showed that the tumor had only slight uptake of 18F-FDG and 11C-choline,and SUVmax was lower than that of 18F-FPA(P<0.01).Conclusion:22RV1 prostate cancer cell line and BALB/c nude mice can be successfully used to develop the tumor-bearing model of prostate cancer in nude mice.18F-FPA has a good imaging effect in 22RV1 type prostate cancer nude mouse model,but it needs to be further compared with other novel imaging agents.
