摘    要:目的:建立1种简单、高效的新生乳鼠大脑皮层神经元原代培养的方法。方法:出生12 h以内的SD乳鼠为研究对象,采用胰酶消化和机械分离法相结合制备细胞悬液;在细胞培养过程中,不加入阿糖胞苷抑制神经胶质细胞生长,而是用2%B27 Neurobasal A medium维持培养。结果:神经元生长良好,形成神经元网络系统;通过神经元免疫组织化学鉴定神经元纯度达90%以上。结论:此方法是获得纯度较高的神经元的1种可靠方法。
