清肺化痰、益气活血两法联合对肺纤维化大鼠血管新生的影响

摘    要:目的比较中药清热化痰、益气活血两法联合运用对肺纤维化(PF)血管新生的影响。方法将77只雄性SPF级W istar大鼠随机分为7组,即假手术组、模型组、中药清肺化痰组、中药益气活血组、两法联合运用组、两法分阶段运用组及氢化可的松组,造模后分别给予相应药液,28 d后取材,链霉亲和素-生物素酶复合物法检测VEGF、Ang-2,进行半定量分级分析,用积分吸光度法计算其阳性面积的IOD值;并观察纤维化肺组织微血管密度的改变。结果模型组肺组织VEGF、Ang-2含量升高,两法联合治疗28 d后,能降低其高表达水平,VEGF、Ang-2分级结果与积分吸光度结果和模型组比较均有显著性差异(P<0.01);28 d时模型组微血管密度明显高于假手术组,提示PF病理性血管新生活跃(P<0.01),各治疗组中,益气活血组、两法联合组与模型组比较有显著性差异(P<0.01)。结论两法联合组兼顾虚、热、痰、瘀致病因素的作用,能降低肺组织中VEGF、Ang-2的分泌,抑制肺组织微血管新生,延缓肺纤维化进程。

Abstract:Objective To compare the influence of the therapy of clearing lung and dissolving phlegm,tonifying qi and activating blood,combined therapy,different stages of medicine administration,4 groups of Chinese herbal drugs and positive control drugs on vessel neogenesis of pulmonary fibrosis.Methods The male Wistar SPF rats(n=77) were randomly divided into 7 groups,including the sham-operauion group(group 1),model group(group 2),group treated with clearing lung and dissolving phlegm therapy(group 3),group with tonifying qi and activating blood therapy(group 4),group with combined therapy(group 5),group treated in different stages(group 6) and group with hydrocortisone(group 7).After 28 days the contents of vascular endothelial growth factor(VEGF) and angiopoietin-2(Ang-2) were detected by avidinbiotin-peroxidasecomplex method(ABC) and semiquantitative grade analysis.The integral optical density value(IODV) was calculated by the integral optical density method.The changes of microvessel density in fibrosis lung tissue were observed.Results In group 2 the contents of VEGF and Ang-2 were increased while decreased 28 days after the treatment with combined therapy.The results of grade and IODV after the treatment had a significant different compared with those of group 2(P<0.01).On 28th day the microvessel density in group 2 was much higher than that of group 1 which indicated that pathological vessel neogenesis of pulmonary fibrosis was activity(P<0.01).In different treatment groups there were significant different among group 4,5 and group 2(P<0.01).Conclusion The therapy combined clearing lung and dissolving phlegm,tonifying qi and activating blood can inhibit the secretion of VEGF and Ang-2 and microvessel neogenesis to delay the development of pulmonary fibrosis.
