MicroRNA-182 在胃癌中的表达及临床意义

目的:探讨微小RNA-182（miRNA-182）在胃癌中的表达及与临床病理特征和预后的关系。方法:收集50例胃癌患者肿瘤组织和非癌胃组织，采用RT-PCR方法检测miRNA-182的相对表达量，并分析其与临床病理资料和预后的相关性。同时，采用RT-PCR方法检测胃癌细胞株和正常胃上皮细胞中miRNA-182的相对表达量。结果:肿瘤组织中miR-182表达水平显著高于非癌胃组织（P＜0.01），人胃癌细胞株中miR-182表达水平明显高于正常胃上皮细胞（P＜0.01）。miR-182的表达水平与胃癌的大小、淋巴结转移、及临床分期呈明显相关（P＜0.05）。miR-182低表达者生存时间明显高于高表达组，差异有显著统计学意义(P＜0．05)。结论:miR-182在胃癌组织和胃癌细胞中表达均上调，且与病情进展和预后相关，可能成为判断胃癌预后的新的分子标记物。

Clinical significance of miRNA-182 in gastric cancer

Objective:To investigate the expression of miRNA-182 in patients with gastric cancer and to analyze the association of the expression with the clinicopathological features and prognosis.Methods:The expression levels of miRNA-182 in 50 tumor tissues and paired normal tissues of gastric cancer were detected by using RT-PCR.The expression of miRNA-182 was also analyzed between gastric cancer cell lines and gastric epithelial cell lines.The correlations between the expression of miRNA-182 and the pathological characteristics and prognosis were analyzed.Results:Expression levels of miRNA-182 were up-regulated in tumor tissues as compared with the paired normal tissues (P＜0.05).A similar pattern was also observed in gastric cancer cell lines.The expression of miRNA-182 was associated with tumor size,lymphatic metastasis,and TNM stages (P＜0.05).The expression of miRNA-182 was correlated with overall survival of gastric cancer (P＜0.05).Conclusion:The expression of miRNA-182 was up-regulated in tumor tissues and cells,and may be a valuable parameter for prognostic evaluation of gastric cancer.
