安胃二号方对胃溃疡大鼠BCL-2及NF-κB的影响

目的:研究安胃二号方对胃溃疡的影响,并探讨其机制。方法:40只wistar大鼠,随机分为空白对照组、病理模型组、奥美拉唑组、安胃二号方组。采用改良Takagi法制作乙酸胃溃疡大鼠模型,治疗13天后测量各组溃疡指数,用免疫组化法检测胃黏膜细胞Bcl-2、NF-κB蛋白的表达。结果:安胃二号方组与其他组比较溃疡指数明显降低(P<0.01),安胃二号方组与奥美拉唑组Bcl-2的表达明显高于模型组(P<0.01),且安胃二号方组高于奥美拉唑组(P<0.05)。安胃二号方组NF-κB的表达明显低于模型组(P<0.01)。结论:安胃二号方能通过增加Bcl-2蛋白表达,抑制NF-κB蛋白的表达,起到对胃溃疡的保护作用,这可能是其促进溃疡愈合机制之一。

The Effects of Anwei Decoction-2 on BCL-2 and NF-κB of Gastric Ulcer of Rats

Objective:To study the protective effects of Anwei Decoction-2(AD-2) on gastric ulcer of rats and its effective mechanism. Methods:Forty healthy wistar rats were randomly divided into model group,AD-2 treatment group,omeprazole treatment group and healthy group.The rat model of gastric ulcer was set up by Takagi's method with some modification.After treating 13d,surveys each group of ulcer index,and the expression of Bcl-2 and NF-κB in the gastric ulcer tissues was examined by immunohistochemical method. Results:AD-2 treatment group could obviously decrease the ulcer index than the others group(P<0.01).Cmpared with the model group,the expression of Bcl-2 was evidently enhanced in the AD-2 treatment group and aomeilazuo treatment group(P<0.01).Significant difference was also noted in the expression level of Bcl-2 between the two treatment groups(P<0.05).Cmpared with the model group,the expression of NF-κB was evidently decrease in the AD-2 treatment group(P<0.01). Conclusion:The results indicated that AD-2 has significantly protection on gastric ulcer.The one of mechanism of its promoting the healing of the gastric ulce may be that the AD-2 can increase the expression of Bcl-2 and reduce NF-κB expression.
