法舒地尔对糖尿病心肌病大鼠左室重构及心力衰竭的影响

摘    要:目的探讨法舒地尔对糖尿痛心肌病大鼠左室重构及心力衰竭的影响。方法成年雄性Wistar大鼠30只,随机分为糖尿病心肌病组(D组,n=10),糖尿病心肌病法舒地尔干预组(T组,n=10),健康大鼠正常对照组(N组,n=10)。T组给予法舒地尔10mg/(kg·d)灌胃,D组和N组给予等量蒸馏水灌胃,每日2次,共12周。分别对3组实验动物进行心脏超声和心功能检测,处死实验动物后采用苦味酸天狼星红染色对左室胶原进行特异染色并计算胶原容积分数(CVF),然后进行观察和比较。结果与N组相比,D组大鼠左室内径扩大,心功能明显下降;T组左室内径增加程度降低,心功能指标改善。苦味酸天狼星红染色显示D组胶原容积分数(CVF)明显增高(P〈0.05);而T组纤维化明显减轻,CVF降低(P〈0.05)。结论法舒地尔对改善糖尿病心肌病大鼠左室重构和心力衰竭有一定的作用。

Effects of Fasudil on Cardiac Left Ventricular Remodeling and Dysfunction in Rats with Experimental Diabetic Cardiomyopathy

Abstract:Objective To investigate the effects of fasudil on cardiac left ventricular remodeling and dysfunction in rats with experimental diabetic cardiomyopathy(DC). Methods DC was induced in male Sprague - Dawley rats by STZ that injected into the intrap- eritoneal. Rats were randomly divided into D group (saline,n=10) ,T groupfasudil 10 rag/( kg · d) ,n=10] for 12 weeks per ga vage. Ten healthful ratsserve as control(N group). Hemodynamics and echocardiographic measurements were obtained at 12 weeks after treatment. The expression and distribution of collagen in LV were investigated with Picric acid - Sirius red staining techniques. Semi - quantitative analysis were used to evaluate the collagen volume fraction(CVF). Results LV cavity dilatation was significantly attenuated in D group. T group partially normalized LV contractile function,which was significantly reduced in D group. LV myocar dial collagen increased and myocardial fibrosis occurred, LV CVF increased significantly, which was partly reversed by fasudil treat ment (P〈0.05). Conclusion Fasduil can decrease the cardiac left ventricular remodeling and dysfunctionand in diabetic cardiomyop athy rats.
