氢质子磁共振波谱活体检测结肠癌耐药性及其分子标志物的探索

摘    要:目的探索氢质子磁共振波谱(1H-magnetic resonance spectroscopy,1H-MRS)活体评价人类结肠癌耐药性及可能分子标志物。材料与方法 8只耐药与8只不耐药人类结肠癌SW480荷瘤裸鼠于肿瘤最大径大于1.5 cm后,分别行1H-MRS检查后,过度麻醉处死荷瘤鼠,切下肿瘤,检测肿瘤的坏死(HE染色)及P-gp、MRP1、PKC、γ-GCSh、γ-GCSl、GSHS、GST蛋白表达(Western blot)。结果耐药组人类结肠癌SW480裸鼠模型Cho峰、Lac峰、Glx1峰、Lip及Ins峰/Cr峰面积较不耐药组人类结肠癌SW480/5-FU裸鼠模型增加,差异有统计学意义(P<0.05)。在HE切片上,组耐药组与不耐药组均见少许坏死,两组组织坏死面积差别无显著性,不耐药组组织间隙较耐药组增大。耐药组PKC、P-gp、MRP1、γ-GCSh、γ-GCSl、GSHS、GST蛋白表达显著高于不耐药组(P<0.05)。Cho峰、Lac峰、Glx1峰、Lip及Ins峰/Cr峰面积与肿瘤耐药相关蛋白表达有明显相关性。结论 Cho峰、Lac峰、Glx1峰及Ins峰/Cr峰面积比有可能作为活体监测结肠癌SW480耐药性的分子标志物。
