| 疏肝健脾方药对非酒精性脂肪性肝病大鼠肝组织SREBP-1c mRNA及蛋白表达的影响 |

| 摘 要： | 目的:观察疏肝健脾方药对非酒精性脂肪性肝病(NAFLD)大鼠肝组织SREBP-1c mRNA及蛋白表达的影响,并探讨其机制。方法:选雄性SPF级SD大鼠55只,随机分为模型组、正常对照组和3个中药干预组,分别为疏肝组、健脾组及合方组。除正常对照组外,各组采用脂肪乳剂灌胃8 w复制大鼠NAFLD模型,同时各药物干预组给予不同药物干预(10 mL/kg),正常对照组和模型组给予等体积蒸馏水灌胃。8 w后处死动物取肝组织,HE染色观察肝脏病理变化;RT-PCR方法检测肝组织SREBP-1c mRNA的表达;免疫组织化学方法检测肝组织SREBP-1c的蛋白表达水平和分布。结果:模型组大鼠肝组织SREBP-1c mRNA表达水平及SREBP-1c蛋白阳性表达率较正常对照组显著升高(P<0.01);各中药干预组大鼠肝组织SREBP-1c mRNA表达水平及SREBP-1c蛋白阳性表达率较模型组显著降低(P<0.05或P<0.01);疏肝组、健脾组大鼠肝组织SREBP-1c mRNA表达率较合方组显著降低(P<0.01)。病理观察显示疏肝、健脾组大鼠肝组织脂肪变最轻。结论:疏肝健脾方药可能是通过下调SREBP-1c mRN...
