摘    要:目的:系统认识近年来上皮钙黏附素系统的基础、临床研究进展。方法:对上皮钙黏附素复合物的分子结构、功能及其调控因素研究进行综合分析讨论,并重点介绍有关研究结果。结果:上皮钙黏附素系统对粘附和维持组织结构完整性发挥重要作用。调控因素主要包括:基因突变,启动子周围甲基化和酪氨酸磷酸化。黏附复合物的表达和功能障碍会导致细胞间粘附丧失、细胞转化和肿瘤发生。结论:钙黏蛋白-连环素复合物不仅仅是黏附分子,对其深入研究有助于阐明肿瘤发生的早期分子改变以及开发肿瘤防治的新策略。

Abstract:Objective:Systemly understanding th e update research progression of E-cadherin adhesion made in basis and clinics.M ethods:This review looks at the gene structure and interaction of the E-cadherin -catenin complex and the factors that appear to regulate E-cadherin expression.R esults:The E-cadherin-catenin complex plays a crucial role in epithelial cell-ce l l adhesion and in the maintenance of tissue architecture.Downregulation of the E -cadherin/catenin complex may occur in several ways amongst which are gene mutat ions,methylation of 5'CpG dinucleotides within the promotor region of E-cadherin ,tyrosine phosphorylation of beta-catenin,Perturbation in the expression or func tion of this complex results in loss of intercellular adhesion,with possible con sequent cell transformation and tumour progression.Conclusion 1:Recent developme nts show that the E-cadherin-catenin complex is more than a'sticky molecular com plex'.further studies may yield greater insight into the early molecular interac tions critical to the initiation and progression of tumours.This should aid the development of novel strategies for both prevention and treatment of cancer.
