铃铛刺花提取物急性毒性及抗疲劳作用研究

摘    要:目的:研究铃铛刺花提取物对实验小鼠的急性毒性作用及抗疲劳作用。方法选择SPF级KM种小鼠为受试对象,采用急性毒性试验测定最大耐受法,采用抗疲劳试验评价其抗疲劳的功效,为其进一步应用的安全性提供理论依据。结果最大耐受量为>120g· kg-1流浸膏,相当于人临床每天口服用量的300倍;与空白对照组比较,铃铛刺花提取物中剂量和高剂量能明显延长小鼠的爬杆时间和小鼠游泳时间,具有统计学意义(P<0.01)。铃铛刺花提取物中剂量和高剂量能显著降低运动后小鼠血乳酸含量,并且能显著提高运动后小鼠肝糖原的含量,具有统计学意义(P<0.01)。铃铛刺花提取物可以将降低运动后小鼠尿素氮的含量,但无统计学意义。结论在本次实验研究条件下铃铛刺花提取物的MTD>120g生药/kg小鼠,实属无毒级,并且铃铛刺花提取物可显著减少运动后乳酸的堆积,提高机体对抗疲劳的能力;可显著提高运动后小鼠肝糖原水平,增加有氧运动糖份供给,从而提高小鼠的抗疲劳作用。

Acute Toxicity and Anti-fatigue Effect of Halimodendron Halodendron Flower Extract in Mice
Abstract:OBJECTIVE To observe the acute toxicity and anti-fatigue effect of Halimodendron halodendron in mice.METHODS Mice of KM species were used to test the maximum tolerance dose of Halimodendron haloden-dron ( Pall.) Voss., evaluate the anti-fatigue effect and provide theoretical basis for safety of its further applica-tion.RESULTS The oral maximum tolerance dose of Halimodendron halodendron ( Pall.) Voss.in mice was >120g· kg-1 ,equal to 300 times than that of human adult′s clinical dose per day.Compared with the control group, mice received high and moderate dosages of Halimodendron halodendron ( Pall.) Voss.flower extract had significant-ly prolonged pole-climbing and swimming time in mice(P<0.01).Moreover,mice received high and moderate dosa-ges of Halimodendron halodendron ( Pall.) Voss.flower extract could significantly lower the concentration of blood lactate and increase the concentration of glycogen in liver.( P<0.01 ) CONCLUSION As demonstrated in this study,the maximum tolerance dose of Halimodendron halodendron ( Pall.) Voss.var.flower extract MTD>120g· kg-1 greater than the regular amount used in clinics, proving that it is non-toxic.Furthermore, the study suggested Halimodendron halodendron ( Pall.) Voss.var.flower extracts can attenuate the aggregation of lactate after exercise, improving the ability against fatigue.Moreover,It could improve the augment the production of the liver glycogen,thus improving the energy supply and the anti-fatigue ability.
