| 摘 要： | 目的建立落新妇苷血药浓度测定方法,研究其在家兔体内的药代动力学。方法家兔耳缘静脉注射10 mg.kg-1落新妇苷,胶束电动毛细管电泳法测定血药浓度,3p97药动学软件分析药动学参数。结果落新妇苷血药浓度在2.05～24.6 mg.L-1范围内呈良好线性,r=0.9993,平均回收率为101.15%,日内、日间RSD均〈6%。落新妇苷在家兔体内的药代动力学过程符合二室开放模型。结论本法简便、快速、准确、干扰小,适用于落新妇苷血药浓度测定,为研究其在人体药代动力学及制定给药方案提供方法学基础。

| Abstract: | Objective To establish a method to determine astilbin in rabbit plasma and to study its pharma- cokinetics. Methods MECC method was adopted after intravenous injection. The data were processed with software 3p97. Results There was a good linear relationship within the range of 2.05-24.6 mg ·L^-1, r =0. 9993. The aver- age recovery was 101.15%. The RSD of intra-day and inter-day assay was less than 6.0%. Conclusion This meth- od is simple, accurate and sensitive,which can be used for determination of astilbin in plasma. This study contrib- utes methodologically to the study of astilbin in human pharmacokinetics and the design of dosage regimens. |
