高血压大鼠肿瘤坏死因子和一氧化氮/一氧化氮合酶系统相关性的研究

摘    要:目的:探讨肿瘤坏死因子(TNF)和一氧化氮(NO)/一氧化氮合酶(NOS)系统在应激性高血压发病中的作用。方法:以电击大鼠足底结合噪音复合刺激制作慢性应激性高血压大鼠(CSHR)模型,采用特异性放射免疫测定技术和化学比色法,检测CSHR和正常血压Wistar大鼠血清中TNF以及血清、心肌和主动脉血管组织NO及NOS的含量。结果:CSHR的尾动脉收缩压(CASP)明显高于Wistar大鼠(P<0.01)。CSHR的血清TNF明显高于Wistar大鼠(P<0.01),而血清、心肌和主动脉血管组织NO及NOS水平则低于Wistar大鼠(P<0.05)。结论:应激刺激作用下TNF增多和NO/NOS系统功能低下可能是造成慢性应激大鼠血压升高的重要因素。

Abstract:Objective:To investigate the effect of tumor necrcsis factor(TNF) and nitric oxide(NO)/nitric oxide synthase(NOS) system in stress-induced hypertension.Methods:The model of chronic stress-induced hypertensive rats(CSHR) was induced by chronic foot-shock and noise stress in adult male wistar rats.The levels of serum TNF and NO/NOS in serum,myocardium and aortic vascular tissues were measured by chemical colorimetry method and radioimmunoassay in CSHR and normal wistar rats.Results:Caudal arterial systolic pressure(CASP) and the level of serum TNF in CSHR were significantly higher than those in wistar rats(P<0.01,P<0.01).The levels of NO/NOS in serum,myocardium and aortic vascular tissues were significantly lower in CSHR than those in wistar rats(P<0.05).Conclusion:Increasing TNF and decreasing function of NO/NOS system induced by stress stimulus are important factors in the occurance of chronic stress-induced hypertension.
