山莨菪碱、ANP和CGRP对油酸型急性肺损伤家兔的治疗作用

摘    要:目的 研究山莨菪碱(654-2)、心钠素(ANP)、降钙素基因相关肽(CGRP)对油本酸型急性肺损伤(ALI)家兔的治疗作用及其机制。方法 采用油酸型ALI模型,将实验动物随机分为模型组(OA组)、各治疗组和对照组(C组),用右颈外静脉插管法测量不同时间点肺动脉压,放免法检测血浆及支气管肺泡灌洗液BALF)中内皮素-1(ET-1)含量,并同时进行动脉血气分析,肺含水量、干湿比测定及病检,结果 与C组相比,OA组平均肺动脉压升高(P<0.05),动脉血氧分压下降(P<0.01),血浆及BALF中内皮素-1含量增加(P<0.01),肺体比值及肺含水量增加(P<0.010,经治疗后,各组均可不同程度降低肺动脉压(654-2组除外)(P<0.05),提升氧分压(P<0.01),降低血压浆及BALF中内皮素-1含量(P<0.05),减轻肺水肿(P<0.01)。结论 654-2,ANP,CGRP对油酸型ALI家兔有治疗作用,联合应用最佳。

Abstract:AIM To investigate the effect and preliminary mechanism of anisodamine (654 2), atrial natriuretic peptide (ANP) and calcitonin gene related peptide (CGRP) on acute lung injury (ALI) rabbits induced by oleic acid. METHODS The rabbits were randomly divided into 6 groups: oleic acid group (OA group), treatment groups and control group. The mean pulmonary arterial pressures (mPAP) at different time points were detected by using right ventricular vessel technique. The concentrations of endothelin 1 (ET 1) in plasma and bronchio alveolar lavage fluid (BALF) at different time points were continuously analysed by radioimmunoassay. In the meantime, PaO 2 and content of lung water were also observed. RESULTS Compared with that in the control group, mPAP, concentrations of ET 1 in plasma and BALF and content of lung water of the oleic acid group markedly increased ( P <0.05, P <0.01), whereas their PaO 2 decreased ( P <0.01). After treatment, mPAP (except 654 2 group), levels of ET 1 and content of lung water decreased ( P <0.05, P <0.01), though varied from group to group, on the other hand, PaO 2 increased ( P <0.01). CONCLUSION Intravenous infusion of 654 2, ANP, CGRP may have beneficial effects on ALI induced by oleic acid.
