应用组织芯片技术检测GPC3蛋白在肝细胞癌及其它肿瘤中的表达

目的 研究GPC3蛋白在肝细胞癌(hepatocellular carcinoma,HCC)及其他人类常见肿瘤组织中的表达。方法 应用组织芯片及免疫组化技术检测36例HCC组织、33例HCC癌旁组织以及其他人类常见肿瘤组织及其癌旁组织中GPC3蛋白的表达。结果 ①GPC3蛋白在HCC、肺鳞癌、癌旁慢性浅表性胃炎组织中表达，而在胃癌、乳腺癌、卵巢癌、胰腺癌、宫颈癌、胆管细胞癌组织中无表达；②36例HCC组织及33例HCC癌旁组织中GPC3蛋白的表达率分别为66.7% (24/36)，3.0%（1/33），二者之间差异有显著性（P＜0.001）；③HCC组织中GPC3蛋白表达与患者年龄、性别、病理分级无相关性（P＞0.05）。结论 GPC3蛋白在HCC、肺鳞癌组织中表达，而在胃癌、乳腺癌、卵巢癌、胰腺癌、宫颈癌中表达沉默，GPC3是一个值得期待的多种肿瘤特异性标志物。

Determination of GPC3 protein’s expression in hepatocellular carcinoma and other tumors with tissue microarray technique

Objective To study the expression of GPC3 protein’s expression in hepatocellular carcinoma(HCC) and other tumors．Methods HCC tissues(36 cases) ,HCC adjacent tissues(33 cases), other human tumor tissues and tumor adjacent tissues were examined for the expression of GPC3 protein by tissue microarry technique and immunohistochemistry.Results ①Expression of GPC3 protein were found in HCC, lung squamous cell carcinoma，chronic superficial gastritis，while not in gastric carcinoma, colorectal carcinoma, pancreatic carcinoma, breast carcinoma, ovarian carcinoma, cervical carcinoma, hepatocholangio carcinoma;②The positive rate of GPC3 protein in HCC tissues (66.7%) was significantly higher than adjacent tissues (3.1%) (P <0.001); ③There were no relationship of GPC3 protein’s expression with gender, age and histological differentiation. Conclusion GPC3 was over-expressed in HCC and lung squamous cell carcinoma tissues，but not in gastric carcinoma, colorectal carcinoma, pancreatic carcinoma, breast carcinoma, ovarian carcinoma, cervical carcinoma, hepatocholangio carcinoma tissue. It suggests GPC3 is a tumor-specific marker of variety tumors.
