| 摘 要： | 目的研究p53正向细胞凋亡调控因子(PUMA)基因mRNA及其功能蛋白在肝癌组织中的表达及其临床意义。方法用半定量RT-PCR和Western blot方法检测42例肝癌患者的癌组织及其癌旁组织和42肝良性病变组织的PUMA mRNA及其编码蛋白表达。结果癌组织、癌旁组织和肝良性病变PUMA mRNA的表达量分别为0.81±0.07、0.97±0.04和1.76±0.06,PUMA蛋白的表达量分别为0.48±0.19、0.61±0.12和1.11±0.34;肝癌组织及癌旁组织的PUMAmRNA及其蛋白表达量明显低于肝良性病变组织高于(P<0.05)。结论 PUMA基因在肝癌组织中的表达下调,与癌的发生呈负相关。

| Abstract: | Objective To explore the expression of p53 up-regulated modulator of apoptosis(PUMA) gene in hepatocellular carcinoma(HCC).Methods With semi-quantitative RT-PCR and Western blot,the expressions of PUMA mRNA and protein were detected in 42 HCC tissues(group A),tumor-adjacent tissues(group B) and 42 benign liver tissues(group C).Results The expressions of PUMA mRNA in groups of A,B and C were 0.81±0.07,0.97±0.04,and 1.76±0.06,respectively,which of PUMA protein were 0.48±0.19,0.61±0.12 and 1.11±0.34.The expressions of PUMA mRNA and its protein were lower in groups of A and B than those in group C(P<0.05).Conclusion The expression of PUMA gene in HCC tissues is down-regulated,which is negatively correlated with the development of liver cancer. |
