SATB1促进肝癌增殖与转移能力的初步研究

目的本研究拟观察SATB1在肝癌中表达情况,并在此基础上利用RNA干扰技术对SATB1进行封闭,观察肝癌生长和转移情况。方法运用实时定量PCR方法检测正常肝组织、肝硬化、良性肝肿瘤及原发性肝癌组织中SATB1的表达情况,并通过实时定量PCR及Western blot检测高转移肝癌细胞株HC-CLM3、低转移肝癌细胞株MHCC-97L及正常肝脏细胞株HL-7702中SATB1的表达。运用RNA干扰技术对高转移肝癌细胞株HCCLM3中的SATB1进行干扰并验证干扰效果,干扰前后的细胞株进行MTT、划痕实验及裸鼠体内移植瘤实验,观察SATB1对高转移肝癌细胞株HCCLM3增殖及转移能力的影响。结果原发性肝癌组织中的SATB1的mRNA水平明显高于正常肝组织、肝硬化组织及良性肝肿瘤组织,且高转移肝癌细胞株HCCLM3中的SATB1表达水平显著高于正常肝脏细胞株HL-7702及低转移肝癌细胞株MHCC-97L;在HCCLM3细胞株中成功干扰SATB1,干扰后细胞株的增殖及迁移能力均明显降低,裸鼠体内肿瘤增殖能力明显减弱。结论 SATB1在肝癌中显著表达,并有促进肝癌细胞株的增殖和转移的作用。

The study of role of SATB1 on liver cancer proliferation and metastasis in liver cancer

Objective To detect the expression of SATB1 in primary liver cancer and study the role of SATB1 in proliferation and metastasis in highly metastatic cell lingage LM3.Methods The expression of SATB1 were detected in normal liver tissue,cirrhosis,benign liver tumors and primary hepatocellular carcinoma by real-time quantitative PCR,and the expression of SATB1 were tested in highly metastatic hepatoma cell lingage HCCLM3,low metastatic hepatoma cell lingage MHCC-97L and normal liver cell lingage HL-7702 by real-time PCR and Western blot.Then interfered the expression of SATB1 in HCCLM3 by RNA interfere,and verified the interference effect.Both cell lineage before and after interference were applied MTT,scratch test and inoculated in nude mice,to clarify the role in the proliferation and metastasis of HCCLM.Results SATB1 mRNA was elevated apparently in primary liver cancer tissue,compared to normal liver tissue,cirrhosis,benign liver tumors,and SATB1 expression increased in HCCLM3 than in HL-7702.In addition,the proliferation and metastasis of HCCLM3 cell lineage decreased after SATB1 was interfered,while in the nude mice xenografts,proliferation decreased.Conclusions SATB1 could be expressed in liver cancer,and promote the proliferation and metastasis of hepatocellular carcinoma cell line.
