薯蓣丸对化疗4T1 乳腺癌小鼠 T 淋巴细胞亚群的影响

摘    要:目的 评价薯蓣丸对化疗后4T1 乳腺癌小鼠免疫功能的影响,并探讨其作用机制。方法 随机 将32 只BALB/c 小鼠分为对照组、模型组、化疗组、中药组,每组8 只。除对照组外,其余组注射4T1 细胞 悬液复制荷瘤模型。成瘤后第1 天化疗组、中药组小鼠予以多西他赛治疗,1 次/7 d,共21 d ;注射多西他赛 后第2 天对中药组小鼠行中药灌胃,1 次/d,共21 d。治疗完成后处死小鼠取脾脏研磨,用流式细胞仪检测小 鼠脾脏中CD4 +、CD8 +、CD4 + CD25 + Foxp3 +调节性T 细胞(Treg)表达水平。结果 与模型组比较,化疗 组、中药组CD4 +、CD8 +、CD4 + CD25 + Foxp3 + Treg 细胞表达水平降低,CD4 + /CD8 +比值升高(P <0.05); 与化疗组比较,中药组CD4 +细胞表达水平、CD4 + /CD8 +比值升高,CD4 + CD25 + Foxp3 + Treg 细胞表达水 平降低(P <0.05)。结论 薯蓣丸能改善化疗后乳腺癌小鼠免疫功能。

Abstract:Objective To evaluate the effect of Dioscorea pills on the immunity of mice with 4T1 breast cancer after chemotherapy and its mechanism of action. Methods Thirty-two BALB/c mice were randomly divided into a control group, a model group, a chemotherapy group and a chemotherapy plus Chinese medicine group with 8 mice in each group. Except the control group, all the other groups were injected with a 4T1 breast cancer cell suspension to build a tumor-bearing mouse model. The chemotherapy group and the chemotherapy plus Chinese medicine group were treated with Docetaxel from the 1st day after tumor formation, once in 7 days, for 21 days; Dioscorea pills were given to the chemotherapy plus Chinese medicine group by gavage from the 2nd day after Docetaxel injection, once a day, for 21 days. After treatment, the mice were sacrificed and their spleens were taken out and ground, and then flow cytometry was used to detect the levels of CD4+, CD8+, CD4+CD25+Foxp3+ Treg cells. Results Compared with the model group, the levels of CD4+, CD8+ and CD4+CD25+Foxp3+ Treg cells decreased, while CD4+/CD8+ ratio increased in the chemotherapy group and the chemotherapy plus Chinese medicine group, and the differences were significant (P < 0.05). Compared with the chemotherapy group, the levels of CD4+ cells and CD4+/CD8+ ratio increased, while the level of CD4+CD25+Foxp3+ Treg cells decreased in the chemotherapy plus Chinese medicine group, and the differences were significant (P < 0.05). Conclusions Dioscorea pills can improve the immune function of mice with breast cancer after chemotherapy.
