摘    要:目的:比较黄芩单味药材提取物和复方双黄连在肠内菌群中代谢的差异.方法:利用离体实验考察黄芩苷在模拟肠道环境下的代谢产物,并通过给大鼠灌胃给药考察实际肠道内的代谢,运用高效液相色谱法和液质联用技术检测和鉴定单方和复方中黄芩苷的代谢情况.结果:单方及复方中黄芩苷在模拟肠道环境下和大鼠肠道内均代谢为黄芩素,体外消除速率常数k分别为0.062 1和0.069 4h-1,t1/2分别为11.2和9.99h,方差分析后进行双单侧检验,F分别为0.21和0.15,小于理论值5.12(P>0.05).结论:单复方中黄芩苷的体外肠菌代谢差异无显著性,但体内代谢产物存在差别.

Abstract:Objective:To compare metabolism of baicalin in skullcap extracts with that in compound Shuanghuanglian(including Scutellaria baicalesis Georgi, Forsythia suspensa Thunb. Vahl and Lonicera japonica Thunb.) granule in rat intestinal bacteria. Methods: The baicalin metabolisms were investigated in mimic intestinal environment by anaerobic culture in vitro and in the rat intestinal tract in vivo, respectively. The baicalin metabolites were determined by HPLC-MS. Results: Baicalin was hydrolyzed to baicaline both in vitro and in vivo. The elimination rate constant (k) of baicalin in skulicap extract and in compound Shuanghuanglian were 0. 0621h~(-1) and 0.0694h~(-1), respectively, t_(1/2)11.2h and 9.99h, respectively, and F value 0.21 and 0.15, respectively, which was lower than the theoretical value of 5.12 (P>0.05). Conclusion : No significant difference in the metabolism rate of baicalin between two resources was found in vitro. However, the baicalin metabolites in compound Shuanghuanglian were different from those in skullcap extract in vivo.
