摘    要:目的:建立鼻咽癌(nasopharyngeal carcinoma,NPC)蛋白质表达谱,为NPC蛋白质组学研究奠定基础。方法:应用激光捕获显微切割(laser capture microdissection,LCM)技术从NPC组织纯化NPC细胞,应用二维凝胶电泳(2-DE)分离LCM 纯化NPC细胞的蛋白质,基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)鉴定蛋白质,利用生物信息学资源构建NPC的2-DE蛋白质组数据库。 结果:建立了NPC蛋白质2-DE参考图谱,2-DE图谱共显示(1 312±30)个蛋白质点,共鉴定了427个蛋白质点,包括241个非冗余蛋白质,并构建了NPC的2-DE蛋白质组数据库,这些数据可从本实验室网站(http://www.xyproteomics.org)上进行查询。结论:首次建立了LCM纯化NPC细胞2-DE蛋白质表达谱,为NPC蛋白质组学研究提供了重要的信息和资料。

Abstract:ObjectiveTo establish the protein expression map of nasopharyngeal carcinoma (NPC), and provide a basis for proteomic study of NPC.MethodsLaser capture microdissection (LCM) was used to isolate cancer cells from NPC tissues. Two-dimensional gel electrophoresis(2-DE) was used to separate the total proteins of LCM purified NPC cells. Matrix-assisted laser desorption/ionization-time of flight-mass spectrometry (MALDI-TOF-MS) was performed to identify the proteins, and bioinformatics was used to construct the 2-DE database of NPC proteome. ResultsA 2-DE reference map of NPC was established. On the 2-DE map, a total of (1 312±30) protein spots were detected, and 427 protein spots representing 241 non-redundant proteins were identified. The 2-DE database of NPC proteome was constructed. These data could be accessed at our website (http://www.xyproteomics.org). ConclusionA protein expression profile of LCM purified NPC tissues has been established for the first time, which provides useful information and source for proteomic study of NPC.
