小牛血清去蛋白注射液对大鼠脑缺血再灌注后血脑屏障通透性的影响及机制分析

摘    要:目的:探讨小牛血清去蛋白注射液(DCSI)对大鼠局灶性脑缺血再灌注后血脑屏障通透性的影响及其可能机制。方法:76只SD大鼠随机分为假手术组(SH)、模型组和DCSI高、低剂量组(DCSI-H、DCSI-L组:60、30mg·kg-1)4组,每组19只,给予相应药物10d后,后3组采用血管内栓线阻断法制备大鼠局灶性脑缺血再灌注损伤模型。于脑缺血2h再灌注24h后测定各组大鼠脑组织水含量、伊文思蓝(EB)含量,电镜观察血脑屏障超微结构的改变,并用Westernblot法观察大鼠脑组织细胞间黏附分子(ICAM)-1、基质金属蛋白(MMP)-9的蛋白表达情况。结果:与模型组比较,DCSI组脑组织水含量、EB含量、ICAM-1和MMP-9的蛋白表达水平显著降低(P<0.05或P<0.01);血脑屏障的破坏程度明显减轻。结论:DCSI可能通过降低ICAM-1、MMP-9的蛋白表达发挥其血脑屏障的保护作用。

Abstract:OBJECTIVE: To study the effect of deproteinized calf serum injection (DCSI) on the permeability of blood-brain barrier (BBB) of rats with cerebral ischemic-reperfusion injury and its mechanism. METHODS: 76 SD rats were randomly divided into sham group (SH), model group, DCSI high-dosage group (DCSI-H, 60 mg·kg-1) and DCSI low-dosage groups (DCSI-L, 30 mg·kg-1)(n=19, respectively). 10 d after giving drugs, local cerebral ischemic-reperfusion model was induced by inserting a wire intro artery in the latter three groups. The contents of water and EB in cerebral tissues were measured, the ultrastructure of BBB were observed by electron microscope and the protein expression of ICAM-1 and MMP-9 were evaluated using Western blot analysis. RESULTS: As compared with model group, in DCSI groups the contents of water and EB and the protein expression of ICAM-1 and MMP-9 were significantly decreased (P0.05 or P0.01). The degree of injury of blood-brain barrier was alleviated in DCSI groups. CONCLUSION: DCSI may protect blood brain barrier by reducing the protein expression of ICAM-1 and MMP-9.
