人胎儿不同发育时期睾丸间质细胞的增殖与凋亡

摘    要:目的:观察不同胎龄人胎儿睾丸间质细胞增殖与凋亡的特征和规律,探讨二者在睾丸发生中的意义。方法:利用SP免疫细胞化学方法和改进的石蜡切片原位末段标记法(TUNEL法)观察了人胎儿12~32周龄睾丸间质细胞增殖与凋亡阳性细胞表达率。结果:细胞凋亡在胎儿16~32周龄睾丸间质细胞呈阳性表达,16~20周为表达高峰,阳性细胞定位于细胞核为桔黄色或棕黄色;PCNA在胎儿16~20周龄睾丸间质细胞呈阳性表达,16~20周为表达高峰,阳性细胞核为桔黄色或棕黄色,随着胎龄的增长呈逐渐下降趋势(P<0.01)。结论:细胞增殖与凋亡在人胎儿睾丸发育过程中有不同程度的表达;表明细胞增殖与凋亡在睾丸组织发育过程中起着重要的作用。

Abstract:Objective : To explore the relationship between proliferation and apoptosis(APO) in the de- velopment of Leydig cells of testis by observing APO and the expressing features of proliferating cell nuclear antigen(PCNA) in different development stage of human fetus. Methods; The positive rate of APO and PCNA in Leydig cells of human fetus were examined from 12th to 32nd week using TUNEL and SP im-munocytochemistry respectively. Results:APO unfold masculine expression in Leydig cell of human fetus from 16w to 32w,the peak of expression was observed from 16w to 20w, positive cell was orange or brown in nucleus. PCNA unfold masculine expression in Leydig cell of hyman fetus from 16w to 20w, the peak of expression was observed from 16w to 20w,positive nucleus was orange or brown,the number of positive cells decreased gradually with the growth of the fetus (P>0. 01). Conclusion:There were different degree expression of PCNA and APO in Leydig cells. It indicated the important action of PCNA and APO in the development of Leydig cells.
