| 摘 要： | 目的:研究民间治疗糖尿病的药物灰兜巴中粗多糖的最佳提取工艺,为该药材质量和制剂研究奠定基础。方法:以粗多糖总糖含量和粗提取物总固体质量为双重指标,苯酚-硫酸法显色后,分光光度法为检测手段,通过提取方法的筛选后,采用单因素试验和正交试验进行提取工艺的系统考察,确定灰兜巴粗多糖的最佳提取工艺。结果:选择传统的水煮回流提取法作为灰兜巴粗多糖的提取方法,最终确定的提取工艺为灰兜巴药材粗粉适量,水煮回流提取次数为2次,第1次加水10倍量,浸泡1h,回流煎煮2h,第2次加水8倍量,回流煎煮2h;药液浓缩比1∶10~1∶15,醇沉浓度80%,静置时间12h。结论:实验结果表明优选出的提取工艺科学、经济,可用于指导质量研究和制剂生产。
