Apelin对肥胖糖尿病大鼠胰岛功能的影响

摘    要:目的:研究孤独G蛋白偶联受体配体(Apelin)对肥胖糖尿病大鼠胰岛功能的影响。方法:健康8周龄的SD雄性大鼠70只,随机取其中20只予普通饲料喂养,50只给予高脂饲料喂养。喂养12周,制造肥胖大鼠模型。肥胖大鼠成模后,随机取肥胖大鼠30只,予左下腹腔内注射链脲佐菌素(STZ)溶液(45mg/kg),制造糖尿病大鼠模型,注射后第4天采用罗氏血糖仪采尾血,测定血糖〉16.67 mmol/L表示已成模。然后进行静脉注射葡萄糖耐量—胰岛素释放试验:实验组1注射葡萄糖(1g/kg)加Apelin-36(200nmol/kg),实验组2仅注射葡萄糖(1g/kg),在注射0,5,10,30,60min各点抽取大鼠静脉血0.5mL,测定胰岛素水平。并计算各组的胰岛素曲线下面积(IAUC)。结果:1肥胖糖尿病组大鼠的血浆胰岛素水平高于正常对照组(P〈0.01),肥胖组大鼠的血浆胰岛素水平高于肥胖糖尿病组(P〈0.01);2正常对照组、肥胖组和肥胖糖尿病组大鼠经同时注射Apelin-36和葡萄糖后,胰岛素曲线下面积均下降,下降幅度分别为40.0%,52.3%和38.1%。结论:Apelin可以减少肥胖糖尿病大鼠葡萄糖刺激的胰岛素释放,降低肥胖糖尿病大鼠的胰岛功能。

Abstract:Objective:To detect the effects of Apelin on the islet function in obesity diabetes rats. Methods: Seventy male SD rats (8 weeks) were used in this study. Twenty rats were randomly selected to feed common forage (control group), and the other 50 rats were raised by the high-fat diet (obesity group) for 12 weeks. Then, thirty obese rats were used to establish the diabetic model by the left lower intraperitoneal injection of STZ solution (45 mg/kg). Four days after injection, if blood glucose concentration from the rat tail was ~ 16.67 mmol/L determined by Roche glucometer, the diabetic model was established. Then, oral glucose test and insulin releasing test were performed: one group injected glucose (1 g/kg) and Apelin36 (200 nmol/kg), and the other group only glucose injection (1 g/kg), and then draw 0.5 ml blood 0, 5, 10, 30, 60 minutes later after injection to measure the insulin level. The IAUC was calculated. Results: Plasma insulin levels were higher in diabetic group than in control group ( P 〈0.01), and in obesity group than in diabetic group ( P 〈0.01). The decrease of insulin area under the curve in control, obesity, and diabetic groups after Apelin-36 and glucose injection at the same time was 40.0%, 52.3% and 38.1%; respectively. Conclusion: Apelin can reduce insulin release by glucose stimulation and islet function in obesity diabetic rats.
