摘    要:目的:研究蝎毒多肽(scorpion venom peptide,SVP)组分Ⅳ,对正常小鼠骨髓造血集落形成的影响.方法:实验分为6组:空白对照组.细胞因子组,SVPIV低剂量组,SVPⅣ高剂量组,SVPⅣ低剂量+细胞因子组、SVPⅣ高剂量+细胞因子组.采用甲基纤维素半固体培养基培养骨髓造血细胞,在第7,11,14 d检测骨髓集落生成单位(CFU)数.采用瑞氏-吉姆萨染色观察细胞形态.结果:在第7 d,SVPⅣ低剂量组CFU数高于SVPⅣ高剂量组,在第11,14 d,SVPⅣ高剂量组CFU数高于细胞因子组和SVPⅣ低剂量组,与空白对照组相比差异有统计学意义(P<0.05).各时间点SVPⅣ+细胞因子组CFU数均高于其它组,其中在第7 d,SVPⅣ低剂量+细胞因子组达到组间最高值,在第11,14 d时,SVPⅣ高剂量+细胞因子组达到组间最高值,与空白对照组相比差异有统计学意义(P<0.05).细胞形态观察结果显示:除了空白对照组,其余各组细胞类型均以粒-单核细胞为主.结论:SVPⅣ具有促进正常小鼠骨髓造血集落形成的作用.

Abstract:Objective :To investigate the effect of scorpion venom peptide componentiv (SVPIV) on promoting haematopoietic colony formation in normal mice. Methods: Experiment was divided into six groups : blank control group,cytokines group,low dosage SVPⅣ group ,high dosage SVPⅣ group,low dosage SVPⅣ combination with cytokines group, high dosage SVP Ⅳ combination with cytokines group . Bone marrow ceils were cultured with methylcellulose culture assay, on day 7, 11 and 14, the number of CFU was calculated. Cells were stained by Wright' s-Giemsa' s dye to identify cellular type. Results:on day 7,the number of CFU was higher in low dosage SVPⅣ group than that in high dosage SVPⅣ group,on day 11,14, the number of CFU was much higher in high dosage SVPⅣ group than that in cytokines group and low dosage SVPⅣ group (P 〈 0. 05 compared with blank control group). The number of CFU was remarkably increased in combined groups among six groups. On day 7 ,low dosage SVPivcombination with cytokines group achieved the maximum, On day 11,14, high dosage SVPⅣ group combination with cytokines group achieved the maximum ( P 〈 0.05 compared with blank control group ). The results of observation cell morphology indicated cellular type was granulocyte-monocyte mainly except the normal control group. Conclusion:SVPiv has effect of promoting haematopoietic colony formation in normal mice.
