基于叶绿体matK和rps16基因序列的黄精属药用植物亲缘关系分析

摘    要:目的:开发适用于区分黄精属主要药用植物的DNA条形码序列,分析物种间亲缘关系。方法:设计黄精属专用的matK和rps 16基因扩增引物,扩增得到多花黄精、长梗黄精、黄精、滇黄精、玉竹、湖北黄精的相应序列,比较序列的种内和种间变异、条形码序列间隔,构建系统发育树,分析38份样品的系谱关系。结果:matK基因的种内变异度高于rps 16,rps 16基因的种间变异度高于matK,结合matK和rps 16基因序列可以应用于区分黄精属的主要药用植物。结论:结合matK和rps 16基因序列的信息可以用于区分黄精属的主要药用植物。

Abstract:Objective:To establish suitable DNA barcode for identification of the main medicinal plants of Polygonatum,and analyze the genetic relationship between species of Polygonatum.Methods:Design of amplified primers for matK and rps 16 genes of Polygonatum,and then the corresponding sequences of P.cyrtonema Hua,P.filipes Merr.,Polygonatum sibiricum Red.,P.kingianum Coll.et Hemsl,P.odoratum(MilL)Druce,and P.zanlanscianense Pamp.were amplified.The intraspecific and interspecific variation,barcoding gap were analyzed.Meanwhile,a phylogenetic tree was constructed to analyze the pedigree relationship of 38 plant materials.Results:The intraspecific variation of matK gene was higher than that of rps 16 gene,while,the interspecific variation of rps 16 gene was higher than that of matK gene.The common medicinal plants of Polygonatum could be distinguished by analysis based on combine segment sequences of matK and rps 16.Conclusion:The identification of common medicinal plants of Polygonatum could be analyzed based on the combine segment of matK and rps16 gene sequences.
