| 摘 要： | 目的优选肺结核诊断中理想的痰TB-DNA提取方法。方法选择我院肺科门诊肺结核患者185例,提取其痰液样本,应用煮沸裂解法、酚/氯仿法、以及进口离心柱法进行分析,对比实时荧光PCR结果。结果进口离心柱法下TB-DNA阳性检出率为81.08%(150/185),明显高于煮沸裂解法以及酚/氯仿法,P<0.05。结论在对疑似肺结核患者痰液标本进行检测中,可优先选择进口离心柱法作为TB-DNA的有效提取方法。

| Abstract: | Objective To select ideal in tuberculosis diagnosis sputum TB-DNA extraction method. Method 185 cases with pulmonary outpatient tuberculosis patients choose our hospital, extract sputum samples, the application of boiling pyrolysis method, phenol/chloroform method, column and import the centrifugal method were analyzed, and compared the real-time fluorescent PCR results. Results Under import centrifugal column method TB-DNA positive detection rate was 81.08%（150/185）, significantly higher than the boiling cracking method and phenol/chloroform method, P〈0.05. Conclusions In patients with suspected pulmonary tuberculosis in sputum specimens for testing, can choose imported centrifugal column priority method as effective TB- DNA extraction method. |
