陡脉冲电场对荷瘤BALB/c小鼠癌细胞杀伤效应和抑制作用的实验研究

摘    要:本研究观测了陡脉冲电场对荷瘤BALB/c小鼠癌细胞的杀伤效应和抑制作用。处理后的癌细胞在光镜下表现为细胞核固缩、核碎裂、核溶解 ;在电镜下表现为异染色质增加 ,边集 ,呈团块样改变 ,胞浆肿胀 ,脂滴数量增加 ,质膜破裂 ;核固缩 ,碎裂 ,线粒体肿胀。抑瘤观察中发现治疗组小鼠肿瘤体积明显减小 ,肿瘤生长受到抑制 ,治疗组和对照组间差异有显著性 ;生存期观察中发现经陡脉冲治疗后的荷瘤小鼠生存期明显延长。实验揭示了陡脉冲电场能有效地杀伤癌细胞 ,明显抑制了荷瘤小鼠恶性肿瘤的生长、增殖 ,有着良好的应用前景。

Abstract:The lethal and inhibitory effects on tumor-bearing mice under steep pulsed electric field were investigated in this work. 50 female BALB/c mice were inoculated with the solution of cervical cancer cells line. Then, the histomorphological changes,suppressive curves and survival effects were examined.The histological and antitumor assay study showed that steep pulsed electric field could selectively injure tumor cells and inhibit cell proliferation ability. After treatment,pyknosis,karyoclasis and karyolysis of tumor cells were observed by light and electron mircroscope.The tumor volumes were measured every ofher day after the treatment. In testing group, complete responses (CR) were 9(45%), progressive diseases (PD) were 11(55%),4 died (20%), but in control group, PD was 20, 9 died (45%). The survival period of testing group and control group was averagely 52.05 days and 33.05 days respectively.There was a significant difference in survival curves (P<0.01). These results indicated that steep pulsedelectric field has lethal and inhibitory effects for cervical cancer. It may prolong the survival period of tumor-bearing mice.
