摘    要:目的:探讨原位PCR技术对检测基因组变异的有效性。方法:应用原位PCR检测豫医无毛小鼠皮肤和脾的石蜡组织切片,并以健康昆明小鼠皮肤和脾的石蜡组织切片、PCR反应液不加Taq DNA聚合酶、不加引物和引物不带生物素作为对照。结果:豫医无毛小鼠皮肤和脾石蜡组织切片中检测到清晰的杂交信号,而且杂交信号位于细胞核内;4组对照均未出现杂交信号。结论:原位PCR在基因组变异检测方面实用、有效。

Abstract:Aim:To explore the application of in situ PCR in genomic DNA alteration.Methods:Paraffin slides of the skin and spleen of Yuyi hairless mice were examined by in situ PCR. Paraffin slides of healthy Kunming mice with PCR reaction without Taq DNA polymerase or primers were set as control.Results:Positive signal was detected in the nuclei on the paraffin slides of Yuyi hairless mice,but not in controls.Conclusion:In situ PCR is very effective in detecting genomic DNA variations.
