姜黄素对Aβ_(25-35)诱导的PC12细胞caspase-3、caspase-8和caspase-9表达的影响

摘    要:目的:探讨姜黄素对β淀粉样蛋白25-35(Aβ_(25-35))诱导的大鼠肾上腺嗜铬细胞瘤PC12细胞存活率、乳酸脱氢酶(LDH)释放率及凋亡的影响。方法:以Aβ_(25-35)作用于PC12细胞,分别采用MTT法和LDH试剂盒检测细胞的存活率和LDH释放率;实验随机分为空白对照组、模型组、姜黄素10μmol/L组和姜黄素20μmol/L组,采用流式细胞术及Annexin V-FITC/PI双染法检测凋亡情况,采用比色法和Western blot法检测凋亡相关蛋白胱天蛋白酶(caspase)-3、caspase-8和caspase-9的活性和表达。结果:与模型组比较,姜黄素可显著升高Aβ_(25-35)诱导的PC12细胞存活率,降低LDH释放率和凋亡率(P0.01);同时可显著降低caspase-3、caspase-8和caspase-9的活性和表达(P0.05或P0.01)。结论:姜黄素可显著抑制caspase-3、caspase-8和caspase-9的表达,从而抑制Aβ_(25-35)诱导的PC12细胞凋亡。

Abstract:AIM: To investigate the effects of curcumin on the viability, the lactate dehydrogenase (LDH) release, the apoptosis, and the activity and the expression levels of caspase-3, caspase-8 and caspase-9 of rat adrenal pheochromocytoma PC12 cells induced by β-amyloid protein 25-35 (Aβ25-35). METHODS: The PC12 cells were treated with Aβ25-35. The viability and LDH release rates were measured by MTT assay and LDH kit, respectively. The cells were randomly divided into blank control group, model group, curcumin 10 μmol/L group and curcumin 20 μmol/L group. The apoptotic rates were evaluated by flow cytometry with Annexin V-FITC/PI staining. The activities and expression levels of caspase-3, caspase-8 and caspase-9 were detected by colorimetric method and Western blot analysis. RESULTS: Compared with model group, curcumin significantly increased the viability, and decreased the LDH release rates and the apoptotic rates of the PC12 cells treated with Aβ25-35(P<0.01). Compared with model group, curcumin significantly decreased the activity and expression levels of caspase-3, caspase-8 and caspase-9 (P<0.05 or P<0.01). CONCLUSION: Curcumin inhibits Aβ25-35-induced apoptosis of PC12 cells by inhibiting the expression of caspase-3, caspase-8 and caspase-9.
