摘    要:以沙门氏菌标准菌株(Salmonella enterica DSM 17058 T)为研究对象,用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)对其进行蛋白质指纹图谱分析,进而得到正确的鉴定结果。为了获得重现性良好的质谱图,对沙门氏菌的最佳培养基、最适培养时间以及样品前处理方法进行了优化,从而确立沙门氏菌MALDI-TOF-MS蛋白质指纹图谱标准分析方法。结果表明:乙醇/甲酸处理法所得图谱中的特征峰多,包含更多的蛋白标志物,所得图谱基线平滑,噪音少,信噪比高,是最佳的样品处理方法。以营养丰富的哥伦比亚琼脂为培养基,在24 h时即可达到强峰值信号,质谱图重现性良好,鉴定结果准确,并且相比于其它3种培养基所需培养时间短。

Abstract:Using Salmonella enterica DSM 17058 T as the research model,the protein fingerprint of Salmonella was studied to obtain an accurate identification result based on MALDI-TOF-MS.In order to achieve a good reproducibility of the protein map and establish the standard analysis method by MALDI-TOF-MS,different mediums,culture times and sample handing methods were optimized.The results showed that more characteristic peak of protein with smooth baseline,less noise and higher signal-to-noise ratio were obtained by ethanol/formic acid treatment method.Taking Columbia Agar Base as the culture medium,the protein map with strong signal and good repeatability was acquired after 24h,which took less time compared with other mediums.
