KAI1转移抑制基因在宫颈鳞癌中的表达及临床意义

摘    要:目的采用免疫组织化学方法检测正常宫颈与宫颈鳞癌KAI1蛋白表达水平,以探讨其与宫颈癌侵袭转移的关系。方法采用免疫组织化学SABC法检测石蜡切片KAI1蛋白表达,样本包括正常对照宫颈组织20例,宫颈上皮内瘤变(CIN)组织20例,宫颈癌组织20例。结果KAI1蛋白表达与宫颈组织恶化有关,在正常宫颈组织、CIN及宫颈癌中表达阳性率分别为95%、90%、40%;三组间差异有统计学意义(P〈0.01)。两两比较各组均有统计学意义(P〈0.01)。KAI1表达与CIN分级无关,Ⅰ、Ⅱ、Ⅲ级的阳性表达率无统计学意义(P〉0.05)。KAI1表达与宫颈鳞癌组织分化程度的关系有关,高分化组较低分化组阳性表达率显著降低(P〈0.01)。宫颈癌按临床分期不同期别越早表达率越高(P〈0.05)。KAI1在未转移组中的阳性率显著高于淋巴转移组(P〈0.05)。结论KAI1基因蛋白在宫颈癌组织中表达减少,蛋白表达强度与宫颈癌组织分化、临床分期及转移均有关,提示KAI1变化可能参与宫颈癌的发生发展,对其检测及进一步研究可能会从一定程度上预测宫颈癌侵袭转移潜能,并作为未来分子治疗的靶基因。

Expression of KAI1 metastasis suppressor gene and its clinical significance in cervical carcinoma

Abstract:Objective To study the expression of KAII tumor suppressor protein by immunohistochemistry. The results were analysis combined with the patho-clinical material. Then determine the relationship between KAI1 and invasiveness and metastasis of cervical cancer. Methods Detect the expression of KAI1 metastasis suppressor protein by immunohistochemistry of paraffin slides. The exponent included 20 cases of normal cervical tissues, 20 cases of cervical intraepithelial neoplasia (CIN) and 20 cases of cervical carcinoma. Results The expression of KAI1 protein was related with the tissue differentiation of cervix. The positive rate of it in normal cervial tissue, CIN and cervical carcinoma were respectively 95% , 90% , 40%. There was significant difference among three group ( P 〈0. 01 ) . The expression of KAI1 protein was not related with the grade of CIN (P 〉0. 05) . The expression of KAI1 protein was related with the differentiation and the clinical stages of cervical cancer (P 〈 0. 01 ) . The expression of KAI1 protein was related with metastasis. The positive rate were significant difference in between the non-metastasis and metastasis group ( P 〈 0. 05 ) . Conclusion There was downregulation of KAI1 metastasis suppressor gene in cervical cancer. Expression of KAII protein is related with the differentiation, clinical stages and metastasis of cervical cancer.
