反相高效液相色谱法测定不同产地枳实中柠檬苦素的含量

摘    要:目的建立反相高效液相色谱法测定枳实中柠檬苦素的含量,并比较不同产地枳实中柠檬苦素的含量。方法采用Promosil C18色谱柱(4.6 mm×250 mm,5μm),流动相为乙腈-水(45:55),流速为1.0mL·min-1,柱温:30℃,检测波长:210 nm。结果枳实中柠檬苦素在0.035 60.534 0μg(r=0.999 2)与峰面积呈良好的线性关系。平均加样回收率为103.9%,RSD为2.3%(n=6)。不同产地的16批枳实药材含量测定结果表明,枳实中柠檬苦素的含量在0.480.534 0μg(r=0.999 2)与峰面积呈良好的线性关系。平均加样回收率为103.9%,RSD为2.3%(n=6)。不同产地的16批枳实药材含量测定结果表明,枳实中柠檬苦素的含量在0.482.44 mg·g-1。结论该方法简便、准确、重复性好,可用于枳实中柠檬苦素的含量测定。

Abstract:Objective To establish an RP-HPLC method to determine the content of limonin in Fructus aurantii immaturus. Methods The samples were separated by a Promosil C18 column(4.6 mm×250 mm, 5 μm) with acetonitrile-water(45 :55) as the mobile phase, the fl ow rate was 1.0 mL·min- 1, the column temperature was 30 ℃, the detective wavelength was 210 nm. Results The linearity of limonin was at 0.035 6- 0.534 0 μg(r = 0.999 2). The average recovery of limonin was 103.9%(RSD = 2.3%). The content of limonin in Fructus aurantii immaturus was 0.48- 2.44 mg·g-1. Conclusion The method is rapid, sensitive and accurate, which can be used for the content determination of limonin in Fructus aurantii immaturus.
