Expression of CDHR2 in lung adenocarcinoma and its correlation with prognosis

Objective To investigate the expression of CDHR2 in lung adenocarcinoma(LUAD)and its relationship with clinicopathological features and prognosis,and to analyze the relationship between CDHR2 and tumor immune invasion in LUAD.Methods The relationship between CDHR2 expression and prognosis was explored in TCGA and GEPIA databases.Immunohistochemical staining was used to detect the expression of CDHR2 in 83 cancer tissues and adjacent normal tissues,and its relationship with clinicopathological features and disease-free survival(DFS)was analyzed.The relationship between CDHR2 and tumor immune invasion level of LUAD was analyzed by TIMER and CIBERSORT database.Results In the TCGA dataset,the expression level of CDHR2 in 515 LUAD tissues was significantly higher than that in 59 adjacent normal tissues(P<0.05).GEPIA database found that the high expression of CDHR2 was significantly associated with worse overall survival time(P<0.05).Immunohistochemistry showed that the high expression rate of CDHR2 protein in LUAD tissues was 66.3%(55/83),which was higher than that in adjacent tissues(37.3%,31/83),and the difference was statistically significant(P<0.05).CDHR2 protein expression was related to TNM stage(P<0.05).The median DFS of patients with high CDHR2 expression was 44.6(95%CI:39.5-49.7)months,which was lower than that of patients with low CDHR2 expression57.3(95%CI:49.9-64.8)months],and the difference was statistically significant(P<0.001).Multivariate Cox regression analysis suggested that lymph node metastasis and CDHR2 protein expression could be independent risk factors affecting the prognosis of LUAD patients(P<0.05).CDHR2 expression was significantly correlated with tumor immune cell infiltration in LUAD(P<0.05).Conclusion The expression of CDHR2 was elevated in LUAD tissues,which was related to TNM stage,tumor immune cell infiltration and prognosis.CDHR2 is expected to be a marker for the diagnosis,treatment and prognosis of LUAD.
