新月菱形藻的紫外诱变及优化培养

摘    要:本实验室通过紫外诱变得到一株富含油脂的新月菱形藻BC6,在实验室条件下研究了不同温度、光照强度、pH值、N源含量、NaCl含量等培养条件对BC6生长的影响。研究结果表明:最适培养温度在20 ℃,pH值为7.5,最适光照强度为54 μE/(m2·s),NaNO3的最适添加量为105 mg/L,NaCl最适添加量为27 g/L。在优化培养条件下,最高细胞密度可达1.69×106 /mL,比优化前增大了2.2倍。

Abstract:We obtained a strain of oil-rich Nitzschia closterium (BC6) through UV-induction. We further investigated the effects of different culture conditions such as temperature, irradiance, pH value, salinity and nitrogen concentration on its growth. The OD68o and growth rate were determined every two days. Experimental results show that the optimum culture conditions are temperature of 20 ℃, pH value of 7.5, irradiance of 54 μmol · m2· s-1, NaNO3 concentration of 105 mg ~ L1 and NaC1 concentration of 27 g · L1. Its highest cell density can reach 1.69 ×10^6/mL, 2.2 times higher than that of non-optimization, under these optimal conditions.
