低能量532nm激光对小鼠视网膜损伤的研究

摘    要:目的 探讨低能量532 nm激光是否对小鼠视网膜产生不可逆的损伤.方法检测20、40、80 mV的激光在0.05、1、3 m距离不同持续照射时间内对视网膜的损伤.激光损伤后5~6 d,做视力、对比敏感度和视网膜电图(ERG)检测,并取出眼球做病理学检测.结果损伤时间为1 s的模型中,小鼠的视力、对比敏感度及ERG未见明显改变;延长至10 s和30 s时,小鼠的视力(P=0.000 3,P=0.000 5)、对比敏感度(P=0.003,P=0.000 4)的改变有统计学意义.所有处理组F-ERG b波振幅无明显降低(P>0.05). 结论低能量532 nm激光眼外照射,可以造成视网膜不可逆的损伤;损伤局限在视网膜神经上皮层,不累及色素上皮层.

Abstract:Objective Many eye doctors are opposed using the antipersonnel laser gun and flash light because they believe which could cause permanent retinal damage even the laser energy is lower than 100 mW.So,laboratory evidence is necessary about this.The purpose of this study was to investigate whether the 532nm laser damage the retina under the lower energy of 20-80 mW.Methods532 nm laser with different energies(20,40,80 mW)were used to irradiate the retinas of 28 mice for 1 second,10 and 30 seconds with the irradiating distance of 3 meters,1 meter and 0.05 meters,and 4 mice were no irradiated as control group.The visual functional examinations including vision acuity,contrast sensitivity and electroretinogram(ERG)were performed in 5 days after laser irradiation.Eyeballs of mice were enucleated for the histopathological observation under the light microscope.ResultsNo significant changes were found in vision acuity,contrast sensitivity and ERG in laser irradiation for 1 second group,however,laser exposure lasting 10 and 30 seconds resulted in the significant decrease in acuity and contrast sensitivity(P<0.01).In irradiation for 1 second at the 3 meters group,no evident morphological change of retina was seen in 20 mW irradiation group under the light microscope and thinned only in focal outer nuclear layer(ONL)and inter nuclear layer(INL)in 40 mW group.The whole layer of retina except RPE were destroyed in 80 mW group.Retina remained intact in morphology in 3 meters irradiation group under the energy of 20 mW for 1 second condition,however,the lesion of retinal ONL and INL became serious at the irradiation distance of 1 meter and 0.05 meters.ConclusionThe 532 nm laser with lower energy of 20-80 mW could results in retinal damage except the RPE layer.
