实时荧光定量PCR检测丙型肝炎病毒

摘    要:目的探讨实时荧光定量PCR(FQ—PCR)技术检测丙型肝炎病毒(HCV)RNA载量与抗-HCV及丙氨酸氨基转移酶(ALT)异常的相关性。方法用FQ—PCR检测115份疑似HCV感染的病人血清HCV—RNA,ELISA法检测抗-HCV,全自动生化分析仪测定ALT。结果以80拷贝/ml血清为标准,115份标本中HCV—RNA阳性率为54.8%(63/115);抗-HCV阳性率为53.9%(62/115);ALT异常率为40.9%(47/115)。HCV—RNA载量与抗-HCV(+)例数呈正相关(r=0.986,P〈0.01)。HCV—RNA载量与ALT异常例数(r=0.94,P〈0.01)、含量(r=0.624,P〈0.01)呈正相关。结论HCV—RNA载量升高,抗-HCV阳性率相应增加,ALT高于正常值(40U/L)的异常率和浓度也增高,均呈正相关。FQ—PCR法可作为临床检测丙型肝炎的确诊方法之一。

Abstract:Objective To investigate the correlation between the quantity of HCV-RNA (detected by Real-time Fluorescent Quantitative PCR) and concentrations of anti-HCV and alanine transaminase (ALT). Methods The quantify of HCV-RNA in 115 serum samples were detected by fluorescent quantitative PCR (FQ-PCR), the concentrations of anti-HCV and ALT were detected by Enzyme immunoassay and automatic biochemistry analyzer. Results In 63 of 115 serum samples, the quantity of HCV-RAN was more than 80 copies per milliliter (the positive rate of HCV-RNA was 54.8%); the positive rate of anti-HCV was 53.9%(62/115); the ALT in 40.9 percent of samples (47/115) was abnormal. It was positive correlation between quantity of HCV-RNA and the number of anti-HCV positive samples (r=0.986, P<0.01). It was positive correlation between the quantity of HCV-RNA and the number of samples with abnormal ALT (r=0.94, P<0.01), and the same between quantity of HCV-RNA and content of ALT(r=0.624, P<0.01). Conclusions It is positive correlation between the quantity of HCV-RNA and the positive rate of anti-HCV, abnormal rate of ALT, content of ALT. FQ-PCR can be a method of diagnosing hepatitis C.
