| 摘 要： | 目的 观察凝血酶对人肝癌细胞株HepG_2增殖的影响及其诱导凋亡的作用.方法 体外培养人肝癌细胞HepG_2,MTT法观测不同浓度凝血酶在不同时间点对肝癌细胞增殖的影响,TUENL法检测其诱导凋亡作用.结果 高浓度凝血酶对肝癌细胞增殖有明显抑制作用(P<0.05),当浓度大于100 U/ml作用时间在48 h后,其抑制率最高可达到(39.08±2.32)%,且不随浓度的提高和时间的延长而增加;高浓度凝血酶能够诱导肝癌细胞凋亡,当浓度大于100 U/ml且作用时间大于48 h后,各组凋亡指数无显著性差异(P>0.05).结论 高浓度凝血酶能够明显抑制体外生长的人肝癌细胞株HepG_2并诱导肝癌细胞凋亡,且存在一定时间(48 h内)及浓度(<100 U/m1)依赖性.

| Abstract: | Objective To investigate the effect of thrombin on the proliferation and apoptosis of human hepatocellular carcinoma cell line HepG2. Methods HepG2 cells were treated with thrombin at the doses of 0,25,50,100,150,or 200 U/ml for 24,48 and 72 h respectively. The proliferation of the cells in each groups was measured by MTT assay.
