冻存喉癌组织的傅立叶变换红外光谱研究

摘    要:目的 通过对冻存喉癌组织与癌旁正常组织进行傅立叶变换红外光谱(FTIR)的对比研究.分析两者光谱的特异性,从而在分子水平上揭示喉癌组织的特征,为喉癌的早期诊断提供依据.方法 FTIR检测仪检测冻存喉癌组织及癌旁正常组织,对比分析两种组织的红外光谱表现包括红外光吸收强度、峰位、峰高(1)以及相对峰强等],并结合常规病理检查结果 ,总结喉癌组织及癌旁正常组织的光谱特征.结果 (1)32例正常癌旁组织的红外光谱彼此相似,所有谱带特征峰的平均出现概率为92%,显示出正常冻存组织红外光谱具有固定的特征峰.(2)喉癌组织红外光谱相对癌旁正常组织发生明显改变:①代表核酸中磷酸基团的1084cm-1谱带明显向高波数方向移动;②碳水化合物分子中的C-O基团的伸缩振动谱带由1165 cm-1附近位移至1170 cm-1;③代表脂类的谱带(如1398cm-1、1452cm-1、2869cm-1处)发生明显变化;④蛋白质酰胺Ⅰ谱带向高波数方向移动.(3)较之癌旁正常组织,喉癌组织Ⅰ2925/Ⅰ2889/Ⅰ1647/Ⅰ1452/Ⅰ1539/Ⅰ1452/Ⅰ1452/Ⅰ1398和Ⅰ1084/Ⅰ1398均发生明显改变(P〈0.05或0.01).(4)不同组织学分化程度喉癌组织间相对峰强未见明显差异(P〉0.05).(5)2例癌旁正常组织被红外光谱指认为癌变,与常规病理检查结果 的符合率为97.5%.结论 FTIR可以在分子水平上反映喉癌组织和正常组织的蛋白质、核酸以及脂类等生物大分子在含量、结构、构象上的差异,能够揭示喉癌组织的特征,有望成为辅助喉癌快速诊断的一种新方法 .

The characteristics of Fourier transform infrared spectroscopy in frozen laryngeal carcinoma tissues

Abstract:Objective To determine the features of frozen laryngeal carcinoma and normal laryngeal tissues by Fourier transform infrared spectroscopy (FTIR). Methods FTIR spectroscopic determination was performed in 32 specimens of frozen laryngeal cancerous tissues and pericancerous normal tissues. The FTIR spectra were analyzed with the pathologic diagnosis for the accuracy. Results The normal tissue specimens presented a similar infrared spectra, the average present rate of characteristic peaks was 92%. The frozen laryngeal carcinoma tissues had similar FIR spectra, but which were different from normal tissues in absorption strength and peak position: (1)The peak at 1084 cm^-1 band in laryngeal carcinoma, which represented the nucleic acid the phosphoric acid groups, markedly shifted to the higher wave number; (2) In the carbohydrate members C-O groups and bases expansion vibrational band moved from 1165 cm^-1 to 1170 cm^-1; (3) The fats bands at 1398cm^-1 , 1452cm^-1 and 2869cm^-1 were considerably shifted. 7) Amide I band shifted to higher wave number significantly. Compared with near normal tissues, the ratios of I2925/I2889 and I1647/I1452 and I1539/I1452 and I1452/I13o8 and I1084/I1398 which in the larygeal carcinoma tissues had obvious change. Compared with pathological diagnosis, two benign specimen was classified as the malignant according to FTIR spectra, with a accuracy of 97.5%. Conclusion The study shows that the malignant and normal laryngeal tissues have different features of FTIR spectra, which are mainly due to changes in protein,nucleic acid and phospholipids. FTIR may be a new method for the diagnosis of laryngeal carcinoma in clinical practice.
