摘    要:目的 研究Bmi-1基因对脐血间充质干细胞(MSCs)增殖的影响.方法 培养人脐血MSCs,先利用RNA干扰技术抑制第3和6代MSCs Bmi-1基因的表达,用real-time PCR检测Bmi-1基因的表达,免疫组织化学检测BrdU掺入率来观察脐血MSCs增殖的能力;再通过连续传代至细胞增殖能力下降,利用real-time PCR检测Bmi-1基因在细胞增殖能力下降前后表达量的变化.结果 经过RNA干扰后,Bmi-1基因表达量显著减少;RNA干扰后的48 h、96 h、168 h,Bmi-1转录水平分别下降为正常的(28.67.±10.39)%、(24.46±12.15)%、 (32.68.±9.34)%.脐血MSCs的BrdU掺入率在RNA干扰前为(38.97 ±5.02)%,干扰后显著下降为(12.62 ±2.91)%.此外,当脐血MSCs体外连续传至12~15代时,伴随其增殖能力的下降,表现在Brdu掺入率降至(4.18 ±1.53)%,Bmi-1转录水平也显著下降(P<0.01).结论 RNA干扰抑制脐血MSCs Bmi-1基因表达后,细胞增殖能力明显下降;细胞连续体外传代至增殖能力下降后,Bmi-1基因表达显著下降.说明Bmi-1基因在脐血MSCs的增殖过程中发挥重要作用.
