高糖诱导大鼠视网膜微血管周细胞Ang-2/Tie-2系统表达改变及牛磺酸的防护效应研究

摘    要:目的通过检测牛磺酸对高糖培养大鼠视网膜微血管周细胞(retinal microvascular pericytes,RMPs)中血管生成素-2(Angiopoietin-2,Ang-2)/Tie-2系统表达的影响,探讨其防护糖尿病视损伤的机制。方法高糖培养大鼠RMPs,实验分正常对照组(Con,5mM D-glucose)、低剂量牛磺酸对照组(Con+0.1mM Tau)、中剂量牛磺酸对照组(Con+1mM Tau)、高剂量牛磺酸对照组(Con+10mM Tau)、甘露醇渗透对照组(Con+mannitol)、高糖组(HG,25mM D-glucose)、低剂量牛磺酸干预高糖组(HG+0.1mM Tau)、中剂量牛磺酸干预高糖组(HG+1mM Tau)、高剂量牛磺酸干预高糖组(HG+10mM Tau)、甘露醇高糖对照组(HG+mannitol)。用免疫细胞荧光双标鉴定大鼠RMPs,用荧光定量PCR技术和ELISA技术检测大鼠RMPs中Ang-2/Tie-2系统表达。结果高糖培养后,大鼠RMPs中Ang-2表达明显增加,Tie-2及磷酸化Tie-2明显降低,牛磺酸干预明显降低高糖组Ang-2表达,明显增加高糖组Tie-2及磷酸化Tie-2水平,呈剂量依赖关系。结论牛磺酸经调节Ang-2/Tie-2系统表达抑制糖尿病引起的视损伤。

The Protection Effect of Taurine on Expressions of Ang-2/Tie-2 System in Rat Retinal Microvascular Pericytes Cultured with High Glucose

Abstract:Objective To investigate the protection molecular mechanism of taurine against diabetic retinopathy by observing the expressions of angiopoietin-2(Ang-2)/Tie-2system in rat retinal microvascular pericytes(RMPs)cultured with high glucose.Methods The rat RMPs cultured with or without high glucose were divided into normal control group and high glucose groups,which were detected at different dosage(0.1,1and 10 mM taurine).And Mannitol was used as osmotic control.The morphology was identified by immunocytochemistry.The changes of Ang-2/Tie-2system expression in rat RMPs were studied through the quantitative real-time polymerase chain reaction(qRT-PCR)analysis and ELISA.Results High glucose could significantly increase the expression of Ang-2in rat RMPs.Taurine intervention can significantly inhibit the up-regulation of Ang-2in RMPs induced by high glucose in a dose(0.1,1,and 10mM)-dependent manner(P〈0.05).while in high glucose group,The Tie-2expression and P-Tie-2level were decreased significantly(P〈0.05).And interestingly taurine showed significant increase in Tie-2expression and P-Tie-2levels(P〈0.05).Conclusion High glucose can up-regulate the expression of Ang-2and down-regulate the Tie-2expression and P-Tie-2levels in rat RMPs.And taurine can protect RMPs from the damage of high glucose,througy regulating the Ang-2/Tie-2system expression.
