葡聚糖凝胶柱色谱法测定阿苯达唑纳米脂质体包封率的方法研究

摘    要:目的:考察用葡聚糖凝胶柱色谱法测定阿苯达唑纳米脂质体包封率的影响因素。方法:考察大、小2种不同粒径(100~300、50~150μm)的葡聚糖凝胶对样品脂质体的分离效果;以方法回收率和脂质体分离比为综合指标,以凝胶柱内径与葡聚糖填充高度比(A)、洗脱流速(B)及上样量(C)为因素设计正交试验,筛选测定脂质体包封率的最优条件,并进行验证试验。结果:小粒径的葡聚糖凝胶对样品分离效果更好;优选测定条件为A:1.2:5、B:5mL·min-1、C:0.5mL;验证试验中平均脂质体分离比(包封率)为(91.56±0.98)%,平均方法回收率为(98.61±0.44)%(n=3)。结论:所建立的阿苯达唑纳米脂质体包封率测定方法准确、重现性好。

Abstract:OBJECTIVE:To determine the influential factors of entrapment efficiency of Albendazole nanoliposomes using sephadex gel column chromatography.METHODS:The separation effect of sephadex gel with diameter of 100~300 μm and 50~150 μm on liposome were investigated.With the method recovery and separation ratio of liposomes as index,the optimal condition for the determination of liposome entrapment efficiency was selected by orthogonal experiment using ratio of gel column internal diameter to glucan packing height(A),elution flow rate(B)and sample loading(C)as factors.RESULTS:The samples were well-separated by sephadex gel with small particle size;the optimal conditions were as follows:A 1.2:5,B 5 mL.min-1 and C 0.5 mL.In validation test,the average separation ratio(entrapment efficiency)of liposome was(91.56±0.98)% with average method recovery of(98.61±0.44)%(n=3).CONCLUSION:The method is accurate,precise and reproducible.
