低强度HiFu辐照对体外培养神经母细胞瘤株SK-N-SH的影响及机制初步探讨

摘    要:目的]研究低强度HiFu辐照对SK-N-SH细胞的影响,并初步探讨其作用机制.方法]MTT法检测不同的时间(10~100 s)超声辐照后SK-N-SH细胞的存活率,以及超声辐照40 8对SK-N-SH细胞VCR化疗敏感性的影响;扫描电镜观察超声辐照后细胞超微结构的改变.结果]低强度HiFu对SK-N-SH细胞有细胞毒作用,辐照时间与细胞存活率之间呈现明显的剂量效应关系;超声辐照可增加细胞对长春新碱的化疗敏感性;超声辐照可导致细胞膜超微结构改变.结论]超声辐照不仅可以杀伤SK-N-SH细胞,而且可以增强SK-N-SH细胞对长春新碱的化疗敏感性,其机制可能是通过改变细胞膜超微结构,使细胞对化疗药物通透性增加,从而发挥其杀伤效应.

Abstract:Objective To explore the cyto-toxic effect of high intensity focused ultrasound(HIFU) on SK-N-SH cells, and to explore its possible mechanisms.Methods The survival rates of SK-N-SH cells were assessed by MTT assay after the cells were exposed to the HIFU irradiation of various time periods(10-100s);MTT assay was used to assess the effect of HIFU irradiation on the chemotherapeutic sensitivity of SK-N-SH cells;The alteration on membranes of SK-N-SH cells exposed to HIFU irradiation were analyzed by scanning electron microscope.Results The low intensity HIFU irradiation had cyto-toxicity on SK-N-SH cells, and the time of irradiation showed significant dose-response effect with the survival rate of cell;The chemotherapeutic sensitivity of SK-N-SH cells towards VCR were increased with the HIFU irradiation;Also HIFU irradiation could induce the change of membranes of irradiated cells.Conclusions SK-N-SH cells could be killed by HIFU irradiation through cyto-toxic effect;moreover, the chemotherapeutic sensitivity of SK-N-SH cells towards VCR could be increased by HI-FU irradiation.The concerned mechanism may involve in the change of ultramicrostructures on membranes of irradiated cells, which could increase the permeability of VCR into the neuroblastoma cells to exert its anti-tumor effect.
