地黄饮子对脑缺血再灌注模型大鼠 Bax,Bcl-2,Caspase-3 蛋白表达的影响

摘    要:目的:探讨地黄饮子对脑缺血再灌注模型大鼠Bax蛋白、Bcl-2蛋白、Caspase-3蛋白表达的影响及其保护大脑损伤的机制.方法:SD大鼠60只,随机分为6组,即假手术组、模型组、阳性药尼莫地平组(1 mg·kg-1)和地黄饮子高、中、低(32,16,8g·kg-1)剂量组,每组10只,用药7d后造模.用结扎大鼠双侧颈总动脉方法建立脑缺血再灌注模型,观察地黄饮子对大鼠脑缺血再灌注120 min后脑组织Bax蛋白、Bcl-2蛋白、Caspase-3蛋白的表达的影响.结果:模型组大鼠Bax蛋白表达升高、Bcl-2蛋白表达降低、Caspase-3蛋白表达升高,与假手术组有显著差异(P<0.05),各药物治疗组能显著降低Bax蛋白表达、升高Bcl-2蛋白表达、降低Caspase-3蛋白表达,与模型组有显著差异(P<0.05).结论:地黄饮子能通过下调Bax蛋白、上调Bcl-2蛋白和下调Caspase-3蛋白表达,抑制脑缺血再灌注损伤后细胞凋亡,从而对脑缺血再灌注损伤起到保护作用.

Abstract:Objective:To explore the effect of Dihuang Yinzi on the expression of Bax, Bcl-2 and Caspase-3 proteins and protective cerebral mechanism after cerebral ischemia reperfusion. Method: Sixty SD rats were selected and randomized into six groups: sham group, model group and high, medium, low dose groups of Dihuang Yinzi(32,16,8 g·kg-1) and nimodipine positive drug group(1 mg·kg-1), each involved ten rats. The cerebral ischemia/reperfusion model was established. The expression of Bax, Bcl-2 and Caspase-3 proteins were detected 120 min after cerebral ischemia/reperfusion treated by Dihuang Yinzi. Result: The expression of Bax and Caspase-3 proteins in the models were significantly higher than those in the sham group (P<0.05). The expression of Bcl-2 proteins in the model group were significantly lower than those in the sham group (P<0.05). While the expression of Bax and Caspase-3 proteins in the treatment group were significantly lower than those in the model group (P<0.05),the expression of Bcl-2 proteins in the treatment group were significantly higher than those in the model group (P<0.05). Conclusion: Dihuang Yinzi can upregulate the expression of Bcl-2 protein and downregulate the expression of Bax and Caspase-3 proteins. Dihuang Yinzi may protect the cerebral injury induced by ischemia/reperfusion through inhibiting neuron apoptosis.
