摘    要:建立了同时测定泽兰中咖啡酸和迷迭香酸的高效液相色谱测定方法,并对不同产地和采收时间的14个样品进行了定量分析。用体积分数为30%的乙醇超声提取样品,用Hypersil ODS2(4.6 mm×250 mm,5μm)分析柱;流动相:A:φ(甲酸)=0.75%的乙腈溶液,B:φ(甲酸)=0.75%的水溶液;梯度洗脱条件:0~5 min,V(A)∶V(B)=10∶90;5~70 min,V(A)∶V(B)=10∶90→V(A)∶V(B)=55∶45;检测波长325 nm;流速1 mL/min;柱温30℃。结果表明,产自河北、山东、江苏、江西、广西、湖北、湖南、安徽、甘肃、北京的10个样品中,以河北产泽兰咖啡酸和迷迭香酸质量分数最高;产于北京的5个不同采收时间的样品以8月样品质量分数高。该方法快速灵敏,准确可靠,可望用于泽兰的质量控制。

Abstract:This inveatigation is to develop a analysis method for the simultaneous determination of caffeic acid and rosmarinci acid in Herba Lycopi by HPLC.Fourteen samples from different habitats and collecting time were analyzed.The samples were extracted with 30%(volumetric percentage) ethanol by ultrasonic.The separation was perfomed by a Hypersil ODS2(5 μm,4.6 mm i.d.×250 mm) at a flow rate of 1.0 mL/min with the detection wavelength at UV325 nm.The mobile phase consisted of acetonitrile with formic acid(0.75%)(A) and water with formic acid(0.75%)(B).The gradient profile was that solution A was held 10% for 5min and then linearly from 10% to 55% in 65 min.The injection volume was 20 μL and the column temperature was maintained at 30 ℃.The highest mass fraction of caffeic acid and rosmarinci acid were detected in Herba Lycopi from Hebei among ten habitats of Hebei,Shandong,Jiangsu,Jiangxi,Guangxi,Hubei,Hunan,Anhui,Gansu,Beijing,and the sample collected in August from Beijing showed the highest content among five samples collected from June to October.The method is sensitive and accurate with good repeatability and suitable for quality control of Herba Lycopi.
