摘    要:建立了一种简单、快速测定甜菜糖蜜中棉籽糖的方法。即样品经超声波提取,碱式醋酸铅脱色净化后,采用高效液相色谱-RID检测器直接测定糖蜜中的棉籽糖含量。色谱条件:色谱柱为Agilent Zorbax carbohydrate柱(4.6×250mm,5μm),流动相为乙腈-水(75:25,V:V),流速lmL/min,柱温30℃(2,RID检测。试验结果表明:棉籽糖在0.5mg/mL-10mg/mL范围内线性良好,相关系数为0.9999;加标回收率为89.7%~102.6%,相对标准偏差(RSD)为1.67%-4.36%(n=3)。

Abstract:A method was developed for the determination of raffionse in beet molasses by high performance liquid chromatography. The sample was extracted with ultrasonic, and decolorized with lead subacetate. Then it was separated on a Zorbax carbohydrate column(250×4.6mm, 5μm)at 30℃, which was then eluted with 75% aeetonitrile aqueous soiution at a rate of 1.0mL/Zmin, and the signal was detected by RID. The results showed that the linear range of raffinose was in the range of 0.5 mg/mL - 10mg/mL with a correlation coefficient of 0.9999. The recoveries of raffinose was 89.7%- 102.6%and the relative standard deviations was1.67% -4.36 % (n = 3).
