参黄降糖汤对2型糖尿病大鼠胰岛素抵抗机制及瘦素水平影响研究

摘    要:目的:探讨参黄降糖汤对2型糖尿病大鼠胰岛素抵抗机制及瘦素水平影响。方法:选取清洁级SD雄性大鼠120只依据随机数字表法将其分成六组,对照组(20只)、模型组(20只)、吡格列酮组(20只)、低剂量组(20只)、中剂量组(20只)及高剂量组(20只);大鼠禁食12小时,将配制好的STZ溶液依据60mg/kg注射入大鼠腹腔内,在上述基础上使用高脂高胆固醇饲料继续喂养4周造模;低剂量、中剂量及高剂量组大鼠参黄降糖汤灌胃,剂量分别为0. 7g/kg·d、1. 4g/kg·d、2. 8g/kg·d灌胃,吡格列酮组大鼠给予吡格列酮1mg/kg·d,对照组和模型组使用10ml/d蒸馏水灌胃,每天一次,共进行八周;观察大鼠一般状况,检测六组大鼠空腹血糖(FPG)、甘油三脂(TG)及胆固醇(CHOL)含量,ELISA法检测大鼠血清瘦素及胰岛素含量,计算大鼠胰岛素抵抗指数(HOMA-IR)。结果:中、高剂量组大鼠FPG、TG含量均低于模型组及低剂量组,差异有统计学意义(P<0. 05);中、高剂量组大鼠血清瘦素含量、胰岛素及HOMA-IR低于模型组及低剂量组,差异均有统计学意义(P<0. 05);中、高剂量组大鼠血清瘦素含量、胰岛素及HOMA-IR高于对照组,差异均有统计学意义(P<0. 05)。中、高剂量组与吡格列酮组大鼠FPG、TG、大鼠血清瘦素含量、胰岛素及HOMA-IR无明显差异,差异无统计学意义(P> 0. 05)。结论:参黄降糖汤可明显改善糖尿病大鼠各项症状,调节血清瘦素、胰岛素含量。

Effect of Shenhuang Jiangtang Decoction on Insulin Resistance and Leptin Level for Type 2 Diabetic Rats

Abstract:Objective:To analyze the effect of Shenhuang Jiangtang Decoction on insulin resistance and leptin level for type2 diabetic rats.Methods:100 clean SD male rats were divided into four groups according to the random number table method:controlled group(20rats),diabetic model group(20rats),pioglitazone group(20rats),medium dose group(20rats) and high dose group(20rats),rats fasted for 12 h,the prepared STZ solution based on 60mg/kg injected into the abdominal cavity of rats,continuous injection of 4weeks in high fat feed to continue feeding 4weeks modeling.The rats in the medium dose and the high dose groups were gavage with Shenhuang Jiangtang Decoction at the doses of 1.4g/kg and 2.8g/kg,respectively.The controlled group was given gavage with 30g/kg distilled water once daily for eight weeks.The general condition of the rats was observed.The levels of fasting blood glucose(FPG),triglyceride(TG) and cholesterol(CHOL) in the five groups were detected.The levels of serum leptin and insulin in the rats were measured by ELISA.The resistance index-IR was caculated.Results:The contents of FPG and TG in middle and the high dose groups were lower than those in the model group and the low dose group(P<0.05).The levels of serum leptin,insulin and HOMA-IR in middle-dose and the high-dose groups were higher than those in the controlled group(P<0.05),and the differences were statistically significant(P<0.05).The levels of serum leptin,insulin and HOMA-IR of FPG,TG and rat in the middle and the high dosage groups and the pioglitazone group had no significant difference(P>0.05).Conclusion:Shenhuang Jiangtang Decoction can significantly improve the symptoms of diabetic rats,regulate serum leptin,insulin content.
